摘要 研究了用固相萃取预分离,高效液相色谱法测定饲料用的灌木中植物的色素的方法;饲料用的灌木样品中的色素用Sep Pak C18固相萃取小柱预分离和富集,以WatersNova Pak C18液相色谱柱为固定相,V(甲醇)∶V(异丙醇)=1∶1和水梯度洗脱为流动相分离。方法可同时测定叶黄素、α 胡萝卜素、β 胡萝卜素、叶绿素 a、叶绿素 b、紫黄质、新叶黄素(新黄质)等色素,方法标准回收率为94%~104%,相对标准偏差为1 9%~2 5%。已用于几种饲料用的植物色素的测定。 A high performance liquid chromatography (HPLC) method for the determination of plant pigments in feedingstuff shrubbery was studied. The plant pigments can be extracted from feedingstuff shrubbery with 90% of acetone by oscillating, then subjected to preseparation and enrichment by solid phase extraction with Sep-Pak-C_(18) cartridge. Chlorophyll-a, chlorophyll-b, α-carotene, β-carbotent, lutein, neoxanthin, violaxanthin, pheophorbide and other plant pigments can be separated on a Nova-Pak-C_(18) column (3.9 mm×150 mm), with methanol+isopropyl alcohol(1+1) and water gradient elution as mobile phase at a flow-rate of (0.5 mL/min.) The photodiode array detector was used to monitor the analytes at the maximum wavelength of each pigment. The composition of mobile phase was as follows: A (methanol+isopropyl alcohol (1+1)), B (water): 0 min(V(A)∶V(B)=9∶1); 10 min((V(A)∶V(B)=1∶0)), 28 min(V(A)∶V(B)=1∶0); 30 min(V(A)∶V(B)=9∶1). The recovery of plant pigments is 94%~104%, and the relative standard deviation is 1.9%~2.5%. This method was applied to the determination of pigments in feedingstuff shrubbery with good results.
