摘要
目的 研究亚甲蓝 (MB) /光化学法灭活 RNA包膜病毒的动力力参数条件。方法 以日本脑炎病毒为指示病毒 ,以与 MB最大吸收波长相匹配的单波长点阵式发光二极管为光源 ,将指示病毒与不同浓度的 MB工作液混合后 ,收集不同光照时间和距离的 MB/病毒混合液 ,将其接种于 Vero单层细胞后 ,空班法测定病毒滴度。结果 当光源光照度一定时 ,MB工作浓度、光源光照时间和距离是影响 MB/光化学法灭活病毒的主要动力学参数 ,1.0μg/ m l的 MB工作浓度可在理想的时间内 (≤ 5 m in)完全杀灭一定光照距离范围内 (≤ 2 .5 m )的病毒 ,同时 ,适当地延长光照时间 (≥ 2 5 m in) ,可完全杀灭更远光照距离的病毒 (≤ 3.0 m )。结论 MB/光化学法可在体外安全高效地杀灭 RNA包膜病毒 ;在光源光照度一定的条件下 ,MB工作浓度、光源光照时间和距离均可影响MB/光化学法灭活病毒的作用效果。
OBJECTIVE To study the dynamic parameters of RNA envelope virus inactivation by methylene blue (MB) / photochemistry method. METHODS Matrix of light emitting diodes matching with maximum absorbing wavelength of MB was used as lighting system; Japanese encephalitis virus was used as indicator of inactivating effects of RNA envelope virus. Mixture of MB and virus suspensions treated with different illuminating time and distance was collected and inoculated in Vero single layer cells. And the virus titer was measured by plaque forming tests. RESULTS Under certain luminous intensity, the main dynamic parametersed of MB/photochemistry consist of MB concentration, illuminating time and distance. One microgramme per milliliter (1.0 μg/ml) of MB could completely inactivate virus in 5 min. When the illuminating time increased to 25 min, 1.0 μg/ml MB could completely inactivate virus in 3.0 m illuminating distance. CONCLUSIONS RNA envelope viruses can be effectively inactivated by MB/photochemistry methods. MB concentration, illuminating time and distance can affect the capabilities of virus inactivation by MB/photochemistry methods under certain luminous intensity.
