羊栖菜褐藻糖胶抗凝血活性的研究 被引量:11

ANTICOAGULANT ACTIVITY OF FUCOIDAN FROM THE BROWN SEAWEED SAGARSSUM FUSIFORM

摘要 本文研究了羊栖菜褐藻糖胶的化学组成和抗凝血活性之间的关系。采用热水提取得羊栖菜粗多糖 ,CaCl2 纯化得褐藻糖胶 ,DEAESepharoseCL 6B柱层析与SepharoseCL 6B柱层析对褐藻糖胶进行分级 ,得到F1、F2、F31、F32和F33五个级分 ,均为岩藻糖、半乳糖和甘露糖等糖基组成的杂多糖 ,并含有硫酸酯和糖醛酸以及少量的蛋白质 ,相对分子质量范围 2 .5万~ 95万。采用活化部分凝血活酶时间 (APTT)和凝血酶时间(TT)检测了这 5个级分的抗凝血活性 ,结果显示 ,羊栖菜褐藻糖胶能显著延长APTT的凝血时间 ,而对TT的影响不明显。F1、F31和F32对APTT的影响比较显著 ,而F2、F33和羊栖菜粗多糖的影响较小。研究表明 ,羊栖菜褐藻糖胶主要是通过抑制内源凝血途径而达到抗凝血的效果 ,其抗凝血活性与褐藻糖胶的硫酸基含量成正相关 ,而与相对分子质量和糖醛酸含量无关。 The relationship between the chemical composition of fucoidan separated from Sargassum fusiform and its anticoagulant activity was studied.Fucoidan was extracted from Sargassum fusiform with hot water,and purified with CaCl 2,and then graded into five fractions:F1,F2,F31,32 and F33 by chromatography on a DEAE Sepharose CL-6B column and a Sepharose CL-6B column.The five fractions all contained fucose,mannose and galactose predominantly,also contained sulphate groups,uronic acid and little protein,and MW from 25 000 to 950 000.The activated partial thromboplastin time(APTT)and thrombin time(TT)were measured to evaluate the anticoagulant activity of the five fractions.The results showed that fucoidan prolonged APTT distinctly,but delayed TT little.Fractions F1,F31 and F32 had larger effect on APTT,while F2,F33 and crude polysaccharide had less effect.Anticoagulant activity mainly came from the endogenesis coagulant approach restrained by fucoidan of Sargassum fusiform.There was a positive correlation between anticoagulant activity and sulfate content of fucoidan,while little relation to MW and content of uronic acid.
