摘要
目的 了解烟台市HIV -1流行毒株的流行情况和基因亚型特征。方法 采集了 7份HIV -1感染者的全血分离单个核细胞 (PBMC) ,提取前病毒DNA ,经nested -PCR扩增HIV -1膜蛋白 (env)基因的核酸片段进行扩增 ,并测定和分析了C2 -V3 区及其邻区 2 70个核苷酸序列。结果 7份HIV -1感染者的PBMC样品中扩增到HIV -1env基因片段 ,经序列分析发现 7份样品中有A、B′和E亚型。 5名有偿献血人员均为B′ (泰国B)亚型 ,2名回国劳务人员分别为A、E亚型。结论 烟台市目前存在HIV -1A、B′和E三种亚型毒株 ,感染情况复杂 ,因此应加强对献血者及回国劳务人员等高危人群的检测和控制 ,以防HIV -1各亚型毒株在烟台市的蔓延。
Objective To study the prevalent status and the feature of gene subtype of HIV-1 strain in Yantai. Methods We collected the whole blood of 7 cases infected with HIV-1 and separated the peripheral mononuclear cells(PBMC).After the provirus DNA was extracted and amplified by nested-PCR,270 nucleotides in HIV-1 membrane protein gene(env)C 2-C 3 region and its neighborhood were sequenced and analyzed. Results HIV-1 env gene fragments were successfully amplified among the PMBC of 7 cases infected with HIV-1.The sequence analysis showed there were three HIV-1 subtypes among 7 samples,which were subtypes A、B′and E.5 remunerative blood donors were all classified as subtype B′(Thailand B),2 returned laborers working abroad were A or E separately. Conclusion There exists A、B′and E the three subtype strains infection in Yantai City,because the condition was complicated,it is important to strengthen the surveillance and control of HIV-1 subtype strain infection in high risk populations especially the remunerative blood donors and returned laborers working abroad to prevent its spread in Yantai City.
