摘要
目的 研究蜂胶的聚酰胺柱分离组分C对Fe2 + /半胱氨酸氧化损伤的体外原代培养乳鼠心肌细胞的保护作用。方法 用 10 0 0 μmol/L的Fe2 + 和 5 0 0 μmol/L的半胱氨酸造成心肌细胞的损伤。加入蜂胶组分C (PropolisC)作为保护剂 ,用槲皮素作为阳性对照药。作用 2 4h后 ,用全自动生化分析仪测定培养介质中LDH的含量 ,用MDA和SOD试剂盒测定细胞中的MDA和SOD含量。结果 Fe2 + /半胱氨酸能明显造成心肌细胞氧化性损伤 ,使LDH释放量和MDA生成量增加、SOD活力下降。加入蜂胶组分C和槲皮素预处理后 ,在低剂量时(2 5 μg/ml)即能使培养介质中LDH量和细胞中MDA量下降 ,并能对SOD产生保护作用 (P <0 0 1) ,该作用呈现良好的量效关系。结论 蜂胶组分C可以减轻活性氧自由基对生物膜的破坏和对SOD的损伤 。
Objective To study the protective function of propolis C isolated from propolis chromatographed on a polyamide column, which was observed in freshly isolated cardiac myocyte damaged by FeSO 4/cysteine.Methods Cultured cardiac myocyte were incubated with FeSO 4 (1000μmol/L) and cysteine (500μmol/l) for 24 hours. Propolis C was used as protective agent and quercetin as a positive control. The activity of LDH was determined by Beckman CX5pro machine. The content of MDA and activity of SOD in cell were determined by MDA and SOD kit.Results FeSO 4/cysteine could induce obvious oxidative injure on cultured cardiac myocyte. It was found the activity of LDH in culture medium and the content of MDA increased and activity of SOD decreased. The cultured cardiac myocyte were preconditioned with propolis C or quercetin for 1 hour, then added FeSO 4/cysteine at 37 ℃ for 24 hours. Propolis C and quercetin decrease the production of MDA and the release of LDH with dose dependent effect. They also protect the activity of SOD in low concentration (25μg/ml).Conclusions Propolis C can decrease the damage of cardiac myocyte membrane by radical and injured of SOD. It suggest that propolis C possesses antioxidant activity.
