摘要
目的探讨蜂胶总黄酮(TFP)对病毒性心肌炎小鼠心肌细胞凋亡的影响及其作用机制。方法将32只小鼠随机分为4组各8只:正常对照组腹腔注射无病毒PRM-1640液,30min后腹腔注射0.9%氯化钠溶液0.1mL;模型组注射0.1mL含1×105TCD50的PRM-1640液建立小鼠病毒性心肌炎模型,接种病毒30min后注射0.9%氯化钠溶液0.1mL,qd,共7d;TFP低剂量组造模30min后腹腔注射TFP100mg·kg-1,qd,共7d;TFP高剂量组造模30min后腹腔注射TFP200mg·kg-1,qd,共7d。第8天处死各组存活小鼠,TUNEL法检测心肌凋亡,WesternBlot法检测心肌Caspase-3活性和表达情况。结果蜂胶总黄酮能显著降低病毒性心肌炎小鼠心肌细胞凋亡,下调Caspase-3表达及活性。结论蜂胶总黄酮能显著抑制小鼠病毒性心肌炎心肌细胞凋亡,机制与抑制Caspase-3表达及活性有关。
Objective To study the effects of total flavones of propolis (TFP) on cardiomyocyte apoptosis of viral myocarditis in mice. Methods Established the model of viral myocarditis in mice by intraperitoneal inoculation with Coxsackie virus B3 solution. Animals were divided into normal control, model control, TFP low dose, and TFP high dose groups. Treatment started 30 min after inoculation, and lasted for 7 d. Cardiomyocyte apoptosis was measured by TUNEL. The expressions and activity of caspase-3 was assessed by using Western blot and so forth. Results TFP decreased the cardiomyocyte apoptosis, inhibited the expression and activity of caspase-3. Conclusion TFP could inhibit cardiomyocyte apoptosis of viral myocarditis in mice, by downregulating expression and activity of caspase-3.
