摘要
利用国际上通用的以底物———对羟基苯海因(HPH)为惟一氮源的培养基从69个土壤样品和3个水样中初步分离出238个菌株,然后利用双层琼脂法和微孔快速筛选法对分离到的菌株和实验室保藏的220多株菌进行反复筛选,最终获得D_海因酶阳性菌株54株.用茚三酮显色法及纸层析法从这54株菌中筛选到2株产N_氨甲酰水解酶的阳性菌株,从而获得了可同时产生D_海因酶(DHase)和D_氨甲酰水解酶(DCase)的菌株.
The screening method were described using DL_HPH as the sole nitrogen source to screen positive strains from 69 soil samples, 3 water samples and 220 strains conserved in our lab. Then all the strains screened out were tested by both spot_test and milipore rapid screening method. Finally, 54 DHase positive strains were screened out. And 2 positive strains simultaneously producing both DHase and DCase were also screened out.
