应用荧光光谱和紫外吸收光谱研究了稀土杂多酸盐EuHSiMo10 W2 O40 ·2 5H2 O (EuW2 )与牛血清白蛋白 (BSA)的相互作用 ,探讨了EuW2 对BSA的荧光猝灭过程的猝灭机理 ,测定了不同温度下该结合反应的结合常数、结合位点数、结合热力学参数 ,并依据能量转移理论确定了给体 -受体间的结合距离 ;用同步荧光技术考察了EuW2
The rare-earth salts of heteropoly acid, EuHSiMo 10W 2O 40·25H 2O (EuW 2), have been synthesized and characterized. The binding reaction between EuW 2 and bovine serum albumin (BSA) was investigated by fluorescence and UV-vis absorption spectroscopy. It is proved that the fluorescence quenching of BSA by EuW 2 is a result of the formation of BSA-EuW 2 complex. The binding site number n, apparent binding constant K and corresponding thermodynamic parameters were measured at different temperatures by fluorescence quenching method. The binding distance r between EuW 2 and BSA was obtained according to Frster theory of non-radiation energy transfer. The effect of EuW 2 on the conformation of BSA was analyzed using synchronous fluorescence spectroscopy.
