The therapeutic effects of Nonsteroidal anti-inflammatory drugs(NSAIDs) are related to COX-2 inhibition and their side effects are related to COX-1 inhibition. Therefore, seeking for the highly selective COX-2 inhibitors has been the hot point of NSAIDs research. The molecular basis and structure-activity relationship of selective cox-2 inhibitors were reviewed.
