牛磺酸对家兔缺血/再灌注心肌细胞凋亡的影响

EFFECTS OF TAURINE ON RABBIT CARDIOMYOCYTE APOPTOSIS DURING ISCHEMIA/REPERFUSION INJURY

摘要 目的 :研究牛磺酸 (Tau)对家兔缺血 /再灌注损伤心肌细胞凋亡的影响。方法 :阻断家兔心脏左冠状动脉前降支 4 5min ,再灌注 180min引起心肌缺血 /再灌注损伤 ,在心肌缺血前 5min耳缘静脉注射牛磺酸 (2 0 0mg/kg) ,应用DNA片段原位末端标记法 (TUNEL染色 ) ,DNA凝胶电泳和流式细胞仪 (FCM )观测心肌细胞凋亡。结果 :琼脂糖凝胶电泳显示损伤对照组 (I/R)心肌DNA呈云梯状改变 ,而Tau +I/R组无此改变。与损伤对照组 (I/R)比较 ,Tau +I/R组缺血心肌凋亡细胞明显减少 (TUNEL染色 )。流式细胞仪测定I/R组及Tau +I/R组缺血心肌凋亡率分别为 17.6 6 %± 1.5 4 %和 4 .86 %± 1.2 3%。I/R组的缺血心肌Fas和Bax蛋白表达较非缺血心肌高 (P <0 .0 1) ,Bcl 2 /Bax比例较非缺血心肌低 (P <0 .0 1) ;而在Tau +I/R组 ,Fas和Bax蛋白表达较I/R组的低 (P <0 .0 1) ,Bcl 2 /Bax比例较I/R组高 (P <0 .0 1)。结论 :牛磺酸可减少I/R家兔心肌细胞凋亡 ,其机制与调控凋亡相关基因Fas ,Bax和Bcl Aim: To study the effect of taurine (Tau) on rabbit cardiomyocyte apoptosis during ischemia/reperfusion(I/R) injury. Me-(thods:)Rabbit heart I/R injury was induced by occluding the left anterior descending coronary artery for 45 min and reperfusion for 180 min. taurine (200 mg/kg) was intravenously injected 5 min before heart ischemia. Cardiomyocyte apoptosis was measured by (using) terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling method (TUNEL), flow cytometry (FCM) and DNA agarose gel electrophoresis. Results: DNA ladder pattern of DNA in myocardium was revealed by agarose gel electrophoresis in I/R group while was not found in Tau+I/R group. Apoptotic cardiomyocytes were sparse within ischemic myocardium at risk in Tau+I/R group as compared with that in I/R group (TUNEL stain). Apoptosis rate in ischemic myocardium from I/R and Tau + I/R groups detected by flow cytometry was 17.66%±1.54% and 4.86%±1.23%, respectively. Fas and Bax protein expressions in ischemic myocardium of I/R group were higher than that in nonischemic myocardium group (P<0.01), Bcl-2/Bax ratio in I/R group was lower than that in nonischemic myocardium(P<0.01); while in Tau+I/R group, Fas and Bax protein expressions were lower than that in I/R group(P<0.01), the Bcl-2/Bax ratio was higher than that in I/R group (P<0.01). Conclusion: Taurine reduced apoptosis of myocytes in I/R rabbit heart; its mechanism may involve Fas, Bax and Bcl-2 proteins expression.
