人骨髓间充质干细胞向骨骼肌定向分化的研究 被引量:1

摘要 目的 研究人骨髓间充质干细胞在裸鼠损伤腿部向骨骼肌定向分化。方法 在裸鼠腿部注入无水酒精造成肌肉损伤 ,取培养人骨髓间充质干细胞用荧光染料Dil标记后注入损伤处 ,术后 6d及 12d取损伤部肌肉进行形态观察和免疫荧光标记。结果 在术后 6d可见Dil标记细胞向骨骼肌分化 ,Myoglobin阳性表达。术后12d后可见新生骨骼肌出现典型横纹 ,细胞核MyoD1阳性表达。 结论 在裸鼠损伤肌肉的微环境中 ,人骨髓间充质干细胞可向骨骼肌定向分化 。 Purpose: Detection of skeletal muscle differentiation of human mesenchymal stem cells in nude mice. Methods: Muscle damage model was established by injected 100% ethanol into the nude mice leg. Human mesenchymal stem cells (HMSC) stained with fluorescence (Dil) was injected into the model. On day 6, 12, morphological observation was performed. Results: On day 6, HMSC stained with Dil could differentiated into skeletal muscle and express Myoglobin positively. On day 12, the typical striped muscle was seen in newborn skeletal muscle. The positive expression of MyoD1 was observed in cell nuclear. Conclusions: In the microenvironment of damage nude, HMSC can differentiated into skeletal muscle, in company with the positive expression of Myoglobin and MyoD1.
