摘要
目的 建立高效液相色谱-蒸发光散射检测器法(HPLC-ELSD)测定天冬中天冬皂苷甲含量的方法。方法 采用ODS色谱柱(C18 5μm,250 mm ×4.6 mm,Alltech,美国),以甲醇-水(75:25)为流动相,流速为0.5 mL·min-1。ELSD参数:漂移管温度为105℃,载气流速为1.6 L·min-1。结果 天冬中天冬皂苷甲的线性范围为0.848-3.053 μg(r=0.999 2),平均回收率(n=5)为98.26%(RSD=2.64%)。结论 本方法简便、准确、重现性好,可为评价不同产地的天冬质量提供一定依据。
OBJECTIVE: To develop a content determination method for astacochioside in Radix Asparagi. METHODS: High performance liquid chromatography-evaporative light scattering detector (HPLC-ELSD) was used. RESULTS: The linear range of astacochioside was 0.848 - 3.053 μg(r = 0.999 2), and the average recovery (n = 5) was 98.26% (RSD = 2.64%). CONCLUSION: This method was simple, accurate and reproducible. This reseach provided basis for the evaluation of the quality of Radix Asparagi in different areas.
