摘要
目的对人类博卡病毒的基因组结构、系统进化以及突变规律进行分析。方法利用生物信息学方法如BLAST、CLUSTALW等程序对已经提交到GenBank数据库中的人类博卡病毒全基因组序列进行比对分析和系统进化分析。结果人类博卡病毒基因组短小、结构简单;与其它博卡病毒基因组同源性高,尤其是犬细小病毒;目前Gen- Bank中的5株人类博卡病毒根据进化关系分析可分为两群(CRD2、st2、CZ643和st1组成一群,WLL-1单独成群);统计突变发现人类博卡病毒NS1基因突变频率最小,VP1/2基因突变频率高。结论人类博卡病毒很可能是动物博卡病毒传染给人后的变种;稳定的NS1基因很可能成为开发疫苗和防治药物的重要靶点。
Objective To analyze the genomic structure,phyletic evolution and mutation rules of human bocavirus. Methods Alignment analysis of three bocavirus genomes and five human bocavirus genomes was done by bioinformatics methods such as program BLAST and CLUSTALW.Results The human bocavirus genome with minute and simple structure has high homology with Bovine Parvovirus and Canine Minute virus genomes,particularly the latter.Phyletic evolution analysis of the five human bocaviruses showed that the human bocavirus isolate CRD2,st2,CZ643 and st1 comprises onegroup,and the human bocavirus WLL-1 belongs to the other.At the same time,it was found that the NS1 gene of human bocavirus has less nucleotide substitutions,but VP1/2 genes have more nucleotide substitutions.Conclusion The results suggest that the human bocavirus might be a new variant of bocavirus from animal and stable NS1 gene is probably an important target of vac- cines and drugs for prevention and cure.
