人参皂苷对Luminal型乳腺癌中Fas、FasL表达水平的影响 被引量:6

Study on Effect of Ginsenoside on Expressions of Fas and FasL in Luminal Breast Cancer

摘要 目的探讨人参皂苷对Luminal型乳腺癌中自杀相关因子(Fas)、Fas配体(FasL)表达水平的影响。方法选取SPF级健康雌性SD大鼠72只,随机分为对照组10只及造模组62只。造模组采用二羟甲基丁酸灌胃建立Luminal型乳腺癌模型,对照组予以等量食用油。将造模成功的大鼠随机分为模型组、人参皂苷低、中、高剂量组及他莫昔芬组,各11只。模型组及对照组给予无菌生理盐水5 mL·kg^-1,qd,灌胃;人参皂苷低、中、高剂量组分别给予0.1 mg·mL^-1、0.2 mg·mL^-1、0.4 mg·mL^-1的人参皂苷溶液5 mL·kg^-1,qd,灌胃;他莫昔芬组予以0.6 mg·mL^-1的他莫昔芬溶液5 mL·kg^-1,qd,灌胃。各组大鼠均给药4周。检测比较各组肿瘤体积和瘤重、抑瘤率,肿瘤组织细胞凋亡指数(AI),血清可溶性Fas(sFas)、可溶性FasL(sFasL)含量,以及肿瘤组织Fas、FasL蛋白表达水平。结果与模型组比较,各组大鼠抑瘤率、肿瘤组织AI、血清sFas含量及肿瘤组织Fas蛋白表达水平较高,较高且人参皂苷低剂量组<人参皂苷中剂量组<人参皂苷高剂量组和他莫昔芬组,差异均有统计学意义(均P<0.05)。与模型组比较,各组大鼠肿瘤体积和瘤重、血清sFasL含量、肿瘤组织FasL蛋白表达水平较低,且人参皂苷低剂量组>人参皂苷中剂量组>人参皂苷高剂量组和他莫昔芬组,差异有统计学意义(P<0.05)。结论人参皂苷能有效抑制Luminal型乳腺癌大鼠肿瘤生长,加速肿瘤细胞凋亡,且药物剂量越高作用效果越好,其作用可能与上调Fas蛋白表达、下调FasL蛋白表达有关。 Objective To analyze the effect of ginsenoside on expressions of factors associated suicide(Fas)andFas ligand(FasL)in Luminal breast cancer.Methods A total of 72 clean healthy female Kunming rats were selected,and were randomly divided into control group with 10 rats and model building group with 62 rats.The model building group was established Luminal breast cancer model by gavage administration of dimethylolbutyric acid.The rats in the control group received the same amount of edible oil.The successful rats were randomly divided into model group,ginsenoside low-dose,medium-dose and high-dose groups and tamoxifen group,11 rats in each group.The model group and control group were given stroke-physiological saline solution 5 mL·kg-1,once a day by gavage administration.The ginsenoside low-dose,medium-dose and high-dose groups were given 0.1 mg·mL-1,0.2 mg·mL-1,0.4 mg·mL-1 ginsenoside solution 5 mL·kg-1,once a day by gavage administration.The tamoxifen group was given 0.6 mg·mL-1 tamoxifen solution 5 mL·kg-1,once a day by gavage administration.All rats in each group were treated for 4 weeks.The tumor volume and weight,tumor suppressor rate,apoptotic index(AI)of tumor tissues,serum contents of soluble Fas(sFas),soluble FasL(sFasL),and expressions of Fas and FasL protein in tumor tissues were detected and compared.Results Compared with those of the model group,the tumor suppressor rate,AI of tumor tissues,serum contents of sFas and expression of Fas in tumor tissues were higher,and ginsenoside low-dose group<ginsenoside middle-dose group<ginsenoside high-dose group and tamoxifen group,the differences were all statistically significant(all P<0.05).Compared with those of the model control group,the tumor volume and weight,serum contents of sFasL and expression of FasL in tumor tissues were lower in each group,and ginsenoside low-dose group>ginsenoside middle-dose group>ginsenoside high-dose group and tamoxifen group,the differences were statistically significant(P<0.05).Conclusion Ginsenosidecan effectively inhibit the growth of Luminal breast cancer in rats,accelerate apoptosis of cancer cells,andthe higher the drug dose,the better the effect,and the effect may be related to up-regulation of Fas protein expression and down-regulation of FasL protein expression.
