喜炎平与头孢唑林钠联用对感染小鼠中性粒细胞凋亡的影响 被引量:2

摘要 目的探讨喜炎平与头孢唑林钠联合应用对感染小鼠中性粒细胞凋亡的影响。方法金葡菌感染小鼠随机分为模型组、头孢唑林钠组、喜炎平组、头孢唑林钠+喜炎平组,另选健康小鼠做对照,每组15只。对照组和模型组给予生理盐水,其他各组均给予相应药物,分别于给药第1、4、7天,小鼠心脏采血,分离血浆和中性粒细胞。流式细胞仪检测中性粒细胞凋亡率,ELISA法检测IL-6、IL-10和sFas含量。结果给药第1天,各组中性粒细胞凋亡率无显著差异(P>0.05);与对照组相比,其他4组IL-6、IL-10和sFas水平显著升高(P<0.05),4组之间无差异(P>0.05)。给药第4天和第7天,与对照组相比,模型组凋亡率显著降低(P<0.05),头孢唑林钠组无差异(P>0.05),喜炎平组及头孢唑林钠+喜炎平组显著升高(P<0.05),后者升高更显著(P<0.05),且用药组凋亡率均显著高于模型组(P<0.05);与对照组相比,模型组IL-6、IL-10和sFas水平显著升高(P<0.05),头孢唑林钠组、喜炎平组、头孢唑林钠+喜炎平组IL-6水平均显著降低(P<0.05)、IL-10和sFas水平显著升高(P<0.05),与模型组相比,3个用药组IL-6水平显著降低(P<0.05)、IL-10水平显著升高(P<0.05),且联合用药组显著优于单用药组(P<0.05)。除对照组之外,其他4组间sFas水平无显著差异(P>0.05)。结论喜炎平与头孢唑林钠联合应用可通过降低血浆IL-6、升高IL-10水平,促进感染小鼠中性粒细胞凋亡,减轻炎症反应。 Objective To investigate the effect of xiyanping combined with cafezoin sodium on apoptosis of neutrophils in mice with infection.Methods Mice with staphylococcus aureus infection were randomly divided into model group,cafezoin sodium group,xiyanping group and cafezoin sodium+xiyanping group,healthy mice were selected as control group,15 mice in each of groups.Control group and model group were given normal saline,and other groups were given corresponding drugs.Collected blood from hearts of mice on day 1,4,7 after administration,then separated plasma and neutrophils.Apoptosis of neutrophils was determined by flow cytometry.The level of IL-6,IL-10 and sFas in plasma were determined by ELISA.Results On day 1 after administration,the apoptosis of neutrophils were no difference among all groups(P>0.05);Compared with control group,the level of IL-6,IL-10 and sFas were improved significantly among other groups(P<0.05),and there was no difference among the 4 groups(P>0.05).on day 4 and7 after administration,compared with control group,apoptosis in model group was reduced significantly(P<0.05),in cafezoin sodium group was no difference(P>0.05),in xiyanping group and cafezoin sodium+xiyanping group were increased significantly(P<0.05),and the latter was more significantly higher(P<0.05),and the apoptosis in groups with drugs were significantly higher than model group(P<0.05).And compared with control group,in model group,the level of IL-6,IL-10 and sFas were increased significantly(P<0.05),in cafezoin sodium group,xiyanping group and cafezoin sodium+xiyanping group,the levels of IL-6 were reduced significantly(P<0.05),the levels of IL-10 and sFas were increased significantly(P<0.05).And compared with model group,in three groups with drugs,the levels of IL-6 were decreased significantly(P<0.05),the levels of IL-10 were increased significantly(P<0.05),and combination group was better than single drug group(P<0.05).The levels of sFas were no difference among model,cafezoin sodium,xiyanping and cafezoin sodium+xiyanping group(P>0.05).Conclusion Xiyanping combined with cafezoin sodium may promote the apoptosis of neutrophils and alleviate inflammation of mice with infection by reducing the level of IL-6 and improving the level of IL-10 in plasma.
