摘要
目的:优选元胡止痛方的乙醇提取工艺,为元胡止痛方的醇提工艺及工业化生产提供实验依据。方法:以乙醇浓度、溶剂体积、提取时间及提取次数为考察因素,按正交表L_(9)(3^(4))进行正交设计实验;通过HPLC法同时测定其指标成分延胡索乙素、欧前胡素的含量,以加权法进行评判,优选其最佳醇提工艺。结果:各因素对元胡止痛方乙醇提取效果的影响由大到小的顺序依次为:B>C>A>D;B、C及A起主要作用,其中提取溶剂(B)、提取时间(C)均有极显著性差异(P<0.01);乙醇浓度(A)有显著性差异(P<0.05);而提取次数(D)无统计学意义(P>0.05)。结论:优选出的元胡止痛方的乙醇提取最佳工艺条件为A_(3)B_(3)C_(3)D_(2),即以10倍量80%乙醇回流提取2次,每次3h为最佳。验证结果表明该工艺稳定、可行,可为元胡止痛方的工业化生产提供参考。
Objective:To optimize the ethanol extraction process of Yuanhu Zhitong Recipe and provide experimental basis for the alcohol extraction technology and industrial production of Yuanhu Zhitong Recipe.Methods:Taking ethanol concentration,solvent volume,extraction time and extraction times as investigating factors,orthogonal design experiment was carried out according to orthogonal table L_(9)(3^(4));the content of dl-tetrahydropalmatine and imperatorinwere determined simultaneously by HPLC,Judging by weighting method,optimizing the best alcohol extraction process.Results:The order of the influence of various factors on the ethanol extraction effect of Yuanhu Zhitong Recipe is:B>C>A>D;B,C and A play the main role,among which(B)extraction solvent,(C)the extraction time has extremely significant difference(P<0.01);(A)the ethanol concentration has a significant difference(P<0.05);and(D)the number of extractions is not statistically significant(P>0.05).Conclusion:The optimal technological condition for ethanol extraction of Yuanhu Zhitong Recipe is A_(3)B_(3)C_(3)D_(2),that is,10 times the amount of 80%ethanol is refluxed twice,and each time is 3 hours.The verification results show that the process is stable and feasible,and can provide a reference for the industrial production of Yuanhu Zhitong Recipe.
