Pumpkin was used as vegetables.They are used popularly in the meals or making drinks because pumpkin contains vitamins,fiber,mineral,protein,glucose and lipid.Besides,pumpkin is not only nourishing,but also is precious drugs to cure diseases.In this article,we analyzed some ingredients including heavy metals and NO3-in soil,water and pumpkin samples from households in Loc Thanh Village,Bao Lam District,Lam Dong Province(pumpkin of households grown by using chemical fertilizers and pesticides).These samples showed that some heavy metals such as Cu(Copper),Pb(Plumb),Cd(Cadmium)and As(Arsen)exceeded the allowable limits.However,the pumpkin samples that we grew by using organic fertilizers showed a significant reduction in heavy metal content,within the safety limit according to Vietnamese standards.Thus,the using organic farming methods has resulted in better quality products than using chemical fertilizers and pesticides. 展开更多

Pumpkin is of great economic importance not only as food resources but also as the most widely used rootstock to graft cucurbit crops.Pumpkin rootstock improves salt tolerance of cucumber scion through respiratory burst oxidase homolog protein D(CmRbohD)-mediated reactive oxygen species(ROS)burst,which further enhances Na^(+)export and K^(+)uptake.Rboh D activation requires calcium signaling.However,the underlying mechanism remains largely unknown.In this study,we discovered two Rboh D members from pumpkin involved in the ROS burst at the early stage of salt stress.CmRbohD1 and CmRbohD2 were functionally redundant and double mutation significantly impaired salt tolerance in pumpkin.Overexpression of CmRbohD1 and CmRbohD2 mitigated salinity-induced damage and maintained a relatively low Na+content and high K+content.We screened the potential calcineurin B-like interacting protein kinases(Cm CIPKs)which could bind with CmRbohD1 and CmRbohD2.Our results revealed that two Rboh Ds formed complexes specifically with CmCIPK1,thereby bursting ROS production.Overexpression of CmCIPK1 promoted the early ROS burst under salt stress condition and maintained a relatively balanced Na+/K+homeostasis.Altogether,we proposed a CmCIPK1-CmRbohD1/D2 complex for pumpkin salt stress signaling transduction,which regulates the ROS burst and Na+/K+homeostasis.Our findings offer unprecedented insights into the general mechanism of pumpkin salt tolerance. 展开更多

以94份黑龙江省籽用南瓜种质资源为试材,采用多样性分析、相关分析、主成分分析和聚类分析的方法,结合表型性状进行鉴定评价,研究了这些材料的种质特性,以期筛选出优异的籽用南瓜种质资源,为籽用南瓜的亲本选配、品种改良和种质创新提... 以94份黑龙江省籽用南瓜种质资源为试材,采用多样性分析、相关分析、主成分分析和聚类分析的方法,结合表型性状进行鉴定评价,研究了这些材料的种质特性,以期筛选出优异的籽用南瓜种质资源,为籽用南瓜的亲本选配、品种改良和种质创新提供参考依据。结果表明:这些种质资源6个表型性状的变异系数在0.225%~0.596%,单瓜籽粒数的变异系数最大,蔓长短的变异系数最小;相关分析结果显示6个表型性状大多呈极显著或显著相关;主成分分析得到2个主成分,累计贡献率为61.380%,代表了瓜和籽粒相关性状的主要因素;聚类分析将所有材料分为2个类群,除籽粒大小性状外,类群I中所鉴定的5个表型性状数值均低于类群II;最终筛选出7份表现优异的籽用南瓜种质资源。 展开更多

[Objective] This paper aims to study a new method of extracting pumpkin polysaccharide from pumpkin. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. 日粮添加不同剂量南瓜粉对育肥牛体外模拟瘤胃发酵参数、生长性能及血液指标的影响

本试验旨在探究日粮添加不同剂量南瓜粉对育肥牛瘤胃发酵参数、生长性能及血液指标的影响。在体外模拟试验中,选择6只月龄、体重相近的育肥牛作为瘤胃液供体,对照组采用基础日粮为底物,试验组在基础日粮底物中添加4%、8%、12%的南瓜粉,... 本试验旨在探究日粮添加不同剂量南瓜粉对育肥牛瘤胃发酵参数、生长性能及血液指标的影响。在体外模拟试验中,选择6只月龄、体重相近的育肥牛作为瘤胃液供体,对照组采用基础日粮为底物,试验组在基础日粮底物中添加4%、8%、12%的南瓜粉,在体外模拟瘤胃发酵培养48 h后,测定产气量和瘤胃发酵参数。在动物试验中,选取13月龄、健康状况良好、体型一致的育肥期公牛24头为研究对象,随机分为4组,每组6个重复。对照组饲喂基础日粮,试验1组、试验2组、试验3组分别在基础日粮中添加30、60、90 g/头南瓜粉,试验持续35 d。体外模拟试验结果表明:与对照组相比,体外发酵48 h试验组累计产气量有提高的趋势但无显著差异(P>0.05);试验1、2、3组干物质降解率较对照组分别提高4.41%、6.76%、7.93%(P>0.05)。试验1、2、3组氨态氮(NH_(3)-N)含量较对照组分别提高10.55%、18.92%、16.29%(P<0.01)。动物试验结果表明:试验2组和试验3组育肥终末体重较对照组分别提高3.03%和0.89%(P>0.05)。试验1、2、3组NH_(3)-N含量分别较对照组提高10.55%、18.92%、16.29%(P<0.01)。试验3组丁酸、乙酸比丙酸较对照组分别极显著提高28.03%、7.63%(P<0.01)。饲喂南瓜粉显著增加了育肥牛血常规中血清抗氧化指标尿素氮(BUN)、C-反应蛋白(CRP)分布宽度变异系数(P<0.05)。试验第14天,试验3组血清中BUN含量较对照组提高18.29%(P<0.01)。试验2组和试验3组血清中CRP含量较对照组分别提高54.42%和46.94%(P<0.05)。试验第28天,试验1、2、3组血清中BUN含量较对照组分别提高了38.91%、21.82%、34.55%(P<0.01)。综上所述,日粮添加南瓜粉能够提高育肥牛体外瘤胃发酵特性以及其生长发育、生产性能和血清指标,建议日粮中南瓜粉的适宜添加量为60 g/头。 展开更多

为探究热处理对南瓜籽分离蛋白(pumpkin seed protein isolate,PSPI)的影响,该文研究了PSPI在80、90、100、120℃条件下,分别处理20 min和40 min,PSPI功能和结构的变化。结果表明,适当的热处理可以显著改善PSPI的溶解性、乳化活性、乳... 为探究热处理对南瓜籽分离蛋白(pumpkin seed protein isolate,PSPI)的影响,该文研究了PSPI在80、90、100、120℃条件下,分别处理20 min和40 min,PSPI功能和结构的变化。结果表明,适当的热处理可以显著改善PSPI的溶解性、乳化活性、乳化稳定性和起泡能力(P<0.05)。这4种功能特性分别在90℃(40 min)、80℃(40 min)、100℃(20 min)和90℃(40 min)条件下提高最为显著(P<0.05),分别提高了39.00%、118.75%、88.29%和27.67%。结构特性方面,适宜的热处理条件不仅能够有效地破坏PSPI蛋白的致密结构,还能提高PSPI可溶性蛋白聚集体的粒径。此外,热处理会导致PSPI的表面疏水性降低,内源荧光增强,这可能与蛋白质分子的重新折叠和聚集相关。综上,该研究通过系统分析加热改性对南瓜籽蛋白结构和功能特性的影响,揭示了热处理在改善南瓜籽蛋白品质特性方面的潜力,为其在南瓜籽蛋白的加工与应用提供了理论支持。 展开更多

为了探明叶面喷施硅肥对籽用美洲南瓜白粉病抗性的影响,本试验以感病品种‘M3’为试材,施以外源硅酸钠,测定其白粉病病情指数、叶片蜡质含量、比叶重、茸毛密度、气孔密度、茸毛长度、叶片厚度、栅栏组织、海绵组织等的变化。结果显示:... 为了探明叶面喷施硅肥对籽用美洲南瓜白粉病抗性的影响,本试验以感病品种‘M3’为试材,施以外源硅酸钠,测定其白粉病病情指数、叶片蜡质含量、比叶重、茸毛密度、气孔密度、茸毛长度、叶片厚度、栅栏组织、海绵组织等的变化。结果显示:随着外源硅酸钠浓度的增加,病情指数逐渐降低,在30 mmol/L浓度时达到最低点。叶片蜡质含量和比叶重在不同浓度处理下无显著差异(P<0.05)。叶片茸毛密度和气孔密度在不同浓度处理下有不同程度的变化,且呈现出明显的规律性。茸毛密度与病情指数、叶片厚度、紧密度呈极显著正相关,而与比叶重、蜡质含量呈负相关。叶片厚度、上表皮厚度、栅栏组织和紧密度也与病情指数呈负相关,而与海绵组织和松散度呈正相关。综上,籽用美洲南瓜在喷施外源硅酸钠后,可以减少气孔密度、茸毛密度,增加叶片厚度、茸毛长度、紧密度等,有效改变叶片结构,增加植株抗性,降低白粉病的发病率。 展开更多

未授粉子房离体培养诱导单倍体是遗传育种的重要方式之一。为探索激素诱导下南瓜未授粉子房胚状体的分子机制,以ZJ-13-2(中国南瓜×印度南瓜)为材料,在分化阶段搭配使用生长素和细胞分裂素进行胚状体诱导试验。结果表明,在培养基中... 未授粉子房离体培养诱导单倍体是遗传育种的重要方式之一。为探索激素诱导下南瓜未授粉子房胚状体的分子机制,以ZJ-13-2(中国南瓜×印度南瓜)为材料,在分化阶段搭配使用生长素和细胞分裂素进行胚状体诱导试验。结果表明,在培养基中添加细胞分裂素和生长素(1.0 mg·L^(-1)6-BA+4.0 mg·L^(-1)2,4-D+0.5 mg·L^(-1)NAA)比单独添加细胞分裂素(1.0 mg·L^(-1)6-BA)胚状体诱导率显著提高,分别为32.1%和13.8%。2组胚状体进行转录组测序,分析激素协同作用下影响胚状体形成的差异基因,共筛选出1197个差异表达基因,675个上调基因,522个下调基因。GO和KEGG富集分析结果表明,差异表达基因主要富集在植物激素信号转导、内质网中的蛋白质加工、淀粉和蔗糖代谢与次级代谢物合成途径。经深入分析,其中441个转录因子(TFs)分属于47个不同的TF家族。CRYAA、CRYAB、HSP20、SKP1、BKI1、BZR2等转录因子可能是调控胚状体发生的关键因子,这些转录因子均表现出显著的表达变化,说明它们可能参与了该南瓜品种胚细胞发生过程的调节。HSF家族成员还与其他转录因子(如WRKY、bZIP和MYB)相互作用,共同调控热胁迫响应基因的表达。这种复杂的调控网络不仅在热胁迫响应中发挥关键作用,也为建立南瓜单倍体高效诱导体系提供了可操作的分子标记与基因资源,加速重要性状的固定与育种进程。 展开更多

[Method] Ehrlich's hematoxylin staining-methyl salicylate clearing technique was used for observation and research of the variation of cell structure and chromosomal behavior during pollen mother cell miosis and male gametophyte development of ‘Tianhong' pumpkin. [Result] The meiosis in pollen moth- er cells of pumpkin was simultaneous cytokinesis. In the process of nuclear division, nuclear membrane and nucleolus of pumpkin pollen mother cells gradually disappeared in the metaphase I and reappeared in telophase I , phragmoplast formed between the two generated crescent-shaped nuclei without cell wall, the phragmoplast gradually disappeared in the metaphase II and reappeared in telophase II. Phragmoplast spread outward from the center of spindle during the second division was connected with that formed on the central interface of two nuclei during the first division, cell wall of microspores generated from periphery to center. Most of the tetrads contained four sub-cells while a few contained extra small cells. During the period of uniuclete microspore at periphery, the single nucleolus split into 2-3 or more small nucleoli, mature pollen grain was two-celled. Mononucleate pollen cells were mostly appeared in the flower buds with length of 1.0-2.0 cm, which could be used as an important indicator to collect materials for anther or microspore culture. [Conclusion] This study laid the foundation for research of the cytogenetics of pumpkin. 展开更多

[Result] The optimum recipe for pumpkin sausage was as follows: 100% of pork(fat to lean ratio of 2∶8), 45% of pumpkin particles, 6% of soy protein, 7% of sugar, 3.5% of salt, 1% of spice and 0.5% of monascus red pigment. NaCl处理对南瓜幼苗生长的影响及耐盐机制分析

为探究盐胁迫对南瓜生长发育的生理调控机制,本研究选取耐盐性差异显著的两个南瓜品系(360-3、20-8)为试材,通过幼苗期盐胁迫试验,系统分析其生理响应特征。试验设置0(CK)、200、300 mmol·L^(-1) 3个NaCl浓度梯度,分析盐胁迫对南... 为探究盐胁迫对南瓜生长发育的生理调控机制,本研究选取耐盐性差异显著的两个南瓜品系(360-3、20-8)为试材,通过幼苗期盐胁迫试验,系统分析其生理响应特征。试验设置0(CK)、200、300 mmol·L^(-1) 3个NaCl浓度梯度,分析盐胁迫对南瓜幼苗生长、生理代谢及抗氧化酶活性的影响。结果表明,随着NaCl浓度升高,两份南瓜材料的幼苗株高显著降低,其中20-8的降幅更为显著。叶片相对含水量(RWC)和干鲜质量随盐浓度增加呈下降趋势,但耐盐品系360-3的RWC在盐胁迫下维持稳定,显著优于20-8。盐胁迫加剧细胞膜损伤,两份南瓜材料的电导率随盐浓度升高显著上升。抗氧化酶活性响应中,360-3在300 mmol·L^(-1) NaCl处理下超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性较对照(0 mmol·L^(-1) NaCl)分别显著提高230.91%、284.72%、40.82%,远高于20-8的103.03%、24.96%、22.89%。两份南瓜材料在300 mmol·L^(-1) NaCl处理下的过氧化氢(H_(2)O_(2))含量较对照分别显著升高37.07%和39.95%,表明抗氧化系统能有效缓解氧化损伤。综上,材料20-8耐盐性差,而材料360-3通过更强的保水能力和抗氧化酶激活机制表现出更强的耐盐性。 展开更多

南瓜籽肽改善运动疲劳作用

本研究建立疲劳小鼠模型,探究不同剂量的南瓜籽肽对小鼠疲劳的改善作用。45只小鼠被随机分为5组,分别是正常对照组、模型组、南瓜籽肽低、中和高剂量组。分析南瓜籽肽的氨基酸组成,并探究不同剂量南瓜籽肽干预后,小鼠力竭游泳时间、血... 本研究建立疲劳小鼠模型,探究不同剂量的南瓜籽肽对小鼠疲劳的改善作用。45只小鼠被随机分为5组,分别是正常对照组、模型组、南瓜籽肽低、中和高剂量组。分析南瓜籽肽的氨基酸组成,并探究不同剂量南瓜籽肽干预后,小鼠力竭游泳时间、血清有害物质、能量物质、氧化应激、炎症水平的变化。结果表明,南瓜籽肽由Glu、Leu和Pro等18种氨基酸组成。动物实验表明,与模型组相比,高剂量南瓜籽肽处理后,小鼠游泳时间、肌糖原和肝糖原含量显著增加(P<0.05),血清尿素氮、乳酸、乳酸脱氢酶含量显著降低(P<0.05)。同时,除肝糖原和乳酸脱氢酶含量外,高剂量组对上述的其他指标改善效果显著(P<0.05)优于低剂量和中剂量组。此外,相比于模型组,高剂量南瓜籽肽喂养的小鼠血清和肝脏中抗氧化酶超氧化物歧化酶(SOD,Superoxide dismutase)、谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)和过氧化氢酶(Catalase,CAT)显著增加(P<0.05),血清和肝脏中丙二醛(Malondialdehyde,MDA)显著降低(P<0.05)。对小鼠炎症水平分析发现,高剂量南瓜籽肽组促炎细胞因子白细胞介素-6(Interleukin 6,IL-6)、肿瘤坏死因子-α(Tumor Necrosis Factor-α,TNF-α)和白细胞介素-1β(Interleukin-1β,IL-1β)显著低于(P<0.05)模型组,抗炎细胞因子白细胞介素-4(Interleukin-4,IL-4)和白细胞介素-10(Interleukin-10,IL-10)显著高于(P<0.05)模型组。最后,相关性分析表明Trp和Leu与促炎细胞因子和血清有害物质(乳酸脱氢酶和血清尿素氮)呈显著负相关(P<0.05)。综上,南瓜籽肽具有抗疲劳作用,高剂量南瓜籽肽作用效果最佳。本研究为南瓜籽肽的应用提供理论支撑。 展开更多

The No.601 watermelon (citrullus lanatus) seeds were treated with 25 keV N+ implantation at the dosage of 7.8 ×1016 ions/cm2. After treatment, watermelon seeds were incubated with 380μg/μl pumpkin (Cucubita, maxima Duch) DNA solution at 35 ℃ for 5 hours. By two-generations of selection and resistance screening at seedling stage, one transformed material was selected out, whose rind color is similar to that of the donor pumpkin and whose size of seeds is between that of the donor and the receptor. Using AFLP (amplified fragment length polymorphism) technique, two polymorphic DNA fragments were amplified. This primarily testified that the donor DNA fragments/gene were introduced into the receptor cell and integrated into the genomic DNA of the receptor. 展开更多

The results showed that the liquid-solid ratio was the most important factor in polysaccharides yield, followed the extraction temperature was the least important factor. The optimum microwave assisted extraction co by ndi extraction time, and tions for the highest polysaccharides yield from pumpkin (16.76%-4-0.38%) were obtained by using the response surface methodology with extraction time of 29 min, an extraction temperature of 79 ℃ and a liquid-solid ratio of 22 mL·g^-1. The polysaccharides from pumpkin fruit (PP) were obtained and purified by hot-water extraction, anion-exchange chromatography, and gel column chromatography. The physicochemical properties of PP were determined by gel filtration chromatography, gas chromatography, fourier transform infrared (FTIR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. Results indicated that the molecular weight of PP was about 23 kDa and PP was composed of D-Arabinose, D-Marmose, D-Glucose, and D-Galactose with a molar ratio of 1 : 7.79 : 70.32 : 7.05. FTIR and NMR spectra indicated that PP was the polysaccharide containing pyranose ring. Additionally, PP protected islets cells from streptozotocin (STZ) injury in vitro via increasing the levels of super-oxide dismutase (SOD) and malondialdehyde (MDA) and reducing the production of NO. The experiment of reverse transcriptase-polymerase chain reaction further proved that PP inhibited apoptosis via modulating the expression of Bax/Bcl-2 in STZ-damaged islet cells. In conclusion, PP could be explored as a novel agent for the treatment of diabetes mellitus. 展开更多
