Pumpkin was used as vegetables.They are used popularly in the meals or making drinks because pumpkin contains vitamins,fiber,mineral,protein,glucose and lipid.Besides,pumpkin is not only nourishing,but also is preciou...Pumpkin was used as vegetables.They are used popularly in the meals or making drinks because pumpkin contains vitamins,fiber,mineral,protein,glucose and lipid.Besides,pumpkin is not only nourishing,but also is precious drugs to cure diseases.In this article,we analyzed some ingredients including heavy metals and NO3-in soil,water and pumpkin samples from households in Loc Thanh Village,Bao Lam District,Lam Dong Province(pumpkin of households grown by using chemical fertilizers and pesticides).These samples showed that some heavy metals such as Cu(Copper),Pb(Plumb),Cd(Cadmium)and As(Arsen)exceeded the allowable limits.However,the pumpkin samples that we grew by using organic fertilizers showed a significant reduction in heavy metal content,within the safety limit according to Vietnamese standards.Thus,the using organic farming methods has resulted in better quality products than using chemical fertilizers and pesticides.展开更多
Pumpkin is of great economic importance not only as food resources but also as the most widely used rootstock to graft cucurbit crops.Pumpkin rootstock improves salt tolerance of cucumber scion through respiratory bur...Pumpkin is of great economic importance not only as food resources but also as the most widely used rootstock to graft cucurbit crops.Pumpkin rootstock improves salt tolerance of cucumber scion through respiratory burst oxidase homolog protein D(CmRbohD)-mediated reactive oxygen species(ROS)burst,which further enhances Na^(+)export and K^(+)uptake.Rboh D activation requires calcium signaling.However,the underlying mechanism remains largely unknown.In this study,we discovered two Rboh D members from pumpkin involved in the ROS burst at the early stage of salt stress.CmRbohD1 and CmRbohD2 were functionally redundant and double mutation significantly impaired salt tolerance in pumpkin.Overexpression of CmRbohD1 and CmRbohD2 mitigated salinity-induced damage and maintained a relatively low Na+content and high K+content.We screened the potential calcineurin B-like interacting protein kinases(Cm CIPKs)which could bind with CmRbohD1 and CmRbohD2.Our results revealed that two Rboh Ds formed complexes specifically with CmCIPK1,thereby bursting ROS production.Overexpression of CmCIPK1 promoted the early ROS burst under salt stress condition and maintained a relatively balanced Na+/K+homeostasis.Altogether,we proposed a CmCIPK1-CmRbohD1/D2 complex for pumpkin salt stress signaling transduction,which regulates the ROS burst and Na+/K+homeostasis.Our findings offer unprecedented insights into the general mechanism of pumpkin salt tolerance.展开更多

Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Method] Ehrlich's hematoxyli...[Objective] Pollen mother cell miosis and male gametophyte development of pumpkin were observed in this study, to provide some cytological basis for pumpkin anther or microspore culture. [Method] Ehrlich's hematoxylin staining-methyl salicylate clearing technique was used for observation and research of the variation of cell structure and chromosomal behavior during pollen mother cell miosis and male gametophyte development of ‘Tianhong' pumpkin. [Result] The meiosis in pollen moth- er cells of pumpkin was simultaneous cytokinesis. In the process of nuclear division, nuclear membrane and nucleolus of pumpkin pollen mother cells gradually disappeared in the metaphase I and reappeared in telophase I , phragmoplast formed between the two generated crescent-shaped nuclei without cell wall, the phragmoplast gradually disappeared in the metaphase II and reappeared in telophase II. Phragmoplast spread outward from the center of spindle during the second division was connected with that formed on the central interface of two nuclei during the first division, cell wall of microspores generated from periphery to center. Most of the tetrads contained four sub-cells while a few contained extra small cells. During the period of uniuclete microspore at periphery, the single nucleolus split into 2-3 or more small nucleoli, mature pollen grain was two-celled. Mononucleate pollen cells were mostly appeared in the flower buds with length of 1.0-2.0 cm, which could be used as an important indicator to collect materials for anther or microspore culture. [Conclusion] This study laid the foundation for research of the cytogenetics of pumpkin.展开更多

After treatment, watermelon seeds were incubated with 380μg/μl pumpkin (Cucubita, maxima Duch) DNA solution at 35 ℃ for 5 hours. By two-generations of selection and resistance screening at seedling stage, one transformed material was selected out, whose rind color is similar to that of the donor pumpkin and whose size of seeds is between that of the donor and the receptor. Using AFLP (amplified fragment length polymorphism) technique, two polymorphic DNA fragments were amplified. This primarily testified that the donor DNA fragments/gene were introduced into the receptor cell and integrated into the genomic DNA of the receptor.展开更多

The physicochemical properties of PP were determined by g...The polysaccharides from pumpkin fruit (PP) were obtained and purified by hot-water extraction, anion-exchange chromatography, and gel column chromatography. The physicochemical properties of PP were determined by gel filtration chromatography, gas chromatography, fourier transform infrared (FTIR) spectroscopy, and nuclear magnetic resonance (NMR) spectroscopy. Results indicated that the molecular weight of PP was about 23 kDa and PP was composed of D-Arabinose, D-Marmose, D-Glucose, and D-Galactose with a molar ratio of 1 : 7.79 : 70.32 : 7.05. FTIR and NMR spectra indicated that PP was the polysaccharide containing pyranose ring. Additionally, PP protected islets cells from streptozotocin (STZ) injury in vitro via increasing the levels of super-oxide dismutase (SOD) and malondialdehyde (MDA) and reducing the production of NO. The experiment of reverse transcriptase-polymerase chain reaction further proved that PP inhibited apoptosis via modulating the expression of Bax/Bcl-2 in STZ-damaged islet cells. In conclusion, PP could be explored as a novel agent for the treatment of diabetes mellitus.展开更多
