Cell culture medium was expo...The aim of this study was to investigate the in vitro cytotoxicity of polyphosphoester polymer used as a novel injectable alveolar bone substitutes for controlled delivery of tetracycline. Cell culture medium was exposed to the polymer (0.01-10 mg/mL) for 24 h. The L-929 mouse fibro- blasts were then exposed to the treated cell culture medium for 24 h. Finally, cell viability and growth were assessed by using MTT assay and Alamar Blue assay. No significant cytotoxicity of the polyphosphoester against L-929 mouse fibroblasts was observed at a concentration up to 10 mg/mL (P〉0.05). The two evaluation methods showed no significant differences (P〉0.05). This study suggests that polyphosphoester does not demonstrate any significant toxic effects to cells in vitro and has the potential to be used both as a medical device and as scaffolds in tissue engineering applications.展开更多

Mg-6%Zn-10%β-Ca3(PO4)2 composite was prepared through powder metallurgy methods with different chitosan coatings on its surface. The properties of the chitosan coatings on the surface of Mg-6%Zn-10%β-Ca3(PO4)2 compo...Mg-6%Zn-10%β-Ca3(PO4)2 composite was prepared through powder metallurgy methods with different chitosan coatings on its surface. The properties of the chitosan coatings on the surface of Mg-6%Zn-10%β-Ca3(PO4)2 composite, such as the adhesion ability, the corrosion behavior and the cytotoxicity properties, were investigated, and the microstructure of the chitosan coating was observed by scanning electron microscope(SEM). The results show that chitosan coating improves the corrosion resistance of the magnesium composite specimens significantly. Mg-6%Zn-10%β-Ca3(PO4)2 composite specimens exhibit good corrosion resistance and low p H values in simulated body fluid(SBF) at 37 °C in the immersion test with 7-layer chitosan coating whose relative molecular mass is 30×104 Da. The cytotoxicity tests indicate that Mg-6%Zn-10%β-Ca3(PO4)2 with chitosan coating is nontoxic with a cytotoxicity grade of zero against L-929 cells, which is better than that of uncoated composites.展开更多

The cytotoxic effect of these sponges was evaluated by using MTT assay. The metabolic high performance liquid chromatography fingerprint of I. echinata was also carried out at two wavelengths(254 and 280 nm).Results: Among the sponges tested in this study, the extracts of I. echinata and Dysidea avara possessed the cytotoxic effect on HeLa and PC12 cells. The obtained fractions from high performance liquid chromatography were evaluated for their cytotoxic properties against the cell lines. The isolated fractions did not show significant cytotoxic properties.Conclusions: I. echinata could be considered as a potential extract for chemotherapy.Further investigation is needed to determine the accuracy of mechanism.展开更多
Objective:The present study has characterized dialyzed toxin from non-0157 VTEC E.coli isolates by vero cell toxicity assay and pathogenecity in mice model.Methods:Toxins from non-0157 verocytotoxic Escherichia coli i...Objective:The present study has characterized dialyzed toxin from non-0157 VTEC E.coli isolates by vero cell toxicity assay and pathogenecity in mice model.Methods:Toxins from non-0157 verocytotoxic Escherichia coli isolated from neonatal calves were characterized.Dialyzed toxin from E.coli 026,0111 and 0103 serotypes were prepared and characterized by verocell toxicity assay and pathogenicity in mice model.E.coli 0157:H7 considered as positive control for this study.Results:Cytopathic effects in vero cell line first rounding of vero cells,followed by clumping of cells and finally disintegrated,blackened,shriveled cell line within 16 to 72 hrs.Phenotypic markers such as hind limb paralysis and reddening of tail were prominent in all the toxicated mice.Extensive histopathological study was conducted for multiple organ involvement.Conclusion: Several methods for toxin assay were developed based on biological,immunological and detection of virulence genes related to toxin production but each test has draw back.Therefore,it is likely that future effort will be focused on the development of assay,which is fast,reliable,specific and sensitive methods based on mice model.展开更多

METHODS: Utilizing the method of computer prediction followed by a 4h(51)Cr release assay con...AIM: To identify hepatitis C virus(HCV) core protein epitopes recognized by HLA-A2 restricted cytotoxic T lymphocyte (CTL). METHODS: Utilizing the method of computer prediction followed by a 4h(51)Cr release assay confirmation. RESULTS: The results showed that peripheral blood mononuclear cells (PBMC) obtained from two HLA-A2 positive donors who were infected with HCV could lyse autologous target cells labeled with peptide "ALAHGVRAL (core 150-158)". The rates of specific lysis of the cells from the two donors were 37.5% and 15.8%, respectively. Blocking of the CTL response with anti-CD4 mAb caused no significant decrease of the specific lysis. But blocking of CTL response with anti-CD8 mAb could abolish the lysis. CONCLUSION: The peptide (core 150-158) is the candidate epitope recognized by HLAA2 restricted CTL.展开更多
OBJECTIVE:To collect and screen for ethnopharmacological properties(antileishmanial,antibacterial and brine lethality assays) of medicinal plant Ocimum basilicum from Peshawar region(34.008 latitude and 71.57 altitude...OBJECTIVE:To collect and screen for ethnopharmacological properties(antileishmanial,antibacterial and brine lethality assays) of medicinal plant Ocimum basilicum from Peshawar region(34.008 latitude and 71.57 altitudes).METHODS:In the present study a general antileishmanial activity against Leishmania tropica strain was carried out.The antibacterial potential of the plant was performed against 06 gram positive and 06 gram negative bacteria.Brine shrimp cytotoxicity assay at different concentrations were investigated.RESULTS:The anti-promastigotes profile of the plant showed good antileishmanial activity exhibited LC_(50) value 21.67 μg/mL.The result for gram positive antibacterial activity revealed that the O.basilicum leaves extract possesses significant inhibitory activity at highest two concentrations ranging from20.66 ± 0.31 to 31.86 ± 0.80 for Clostridium perfringens type C and Bacillus subtitilis,respectively,as compared to the gentamycin(27.36 ± 0.55 and21.80 ± 0.72,respectively).For gram negative bacteria good activity was observed.A highest zone of inhibition was recorded for Pseudomonas aeroginosa(28.83 ± 0.28) atthe highest concentration(10 mg/mL).The LC_(50) value obtained for brine shrimp lethality assay was 91.56 μg/mL.CONCLUSION:The herb basil possesses effective cidal activities which make this plant a good candidate for the isolation of antiprotozoal and antibacterial compounds which may lead to the development of novel drug.展开更多
Some novel dicoumarin derivatives, triethylene-glycol dibenzo[5,6] coumarin-3-carboxylate(la), PEG (600) dibenzo[5,6]coumarin-3-carboxylate(1b), triethylene-glycol di[7-(N,N'-diethylamino)]-coumarin-3-carboxy...Some novel dicoumarin derivatives, triethylene-glycol dibenzo[5,6] coumarin-3-carboxylate(la), PEG (600) dibenzo[5,6]coumarin-3-carboxylate(1b), triethylene-glycol di[7-(N,N'-diethylamino)]-coumarin-3-carboxylate(2a), were synthesized. The cytotoxic effect of these compounds, along with benzo[5,6]coumarin-3-carboxylic acid(l) and 7-(N,N'-diethylamino)-coumarin-3-carboxylic acid(2), against the SGC-7901 cell lines were determined by Sulforhodamine B(SRB) assay. The preliminary cytotoxicity screening process revealed that the investigated dicoumarin derivatives induced 50% inhibition of the cell viability of SGC-7901 cells at micromolar concentrations. Compound 2a was proved superior to compound la according to the IC50 values obtained and the agent with PEG moiety has more contribution to cell-killing ability of the molecules than the remaining agents.展开更多
