目的探究LINC01354调控脑胶质瘤细胞增殖、凋亡及焦亡的功能及机制。方法实时荧光定量聚合酶链反应(qRT-PCR)检测人小胶质细胞HMC3和脑胶质瘤细胞U251中LINC01354相对表达量。双荧光素酶报告基因实验验证LINC01354和miR-214-3p的靶向关... 目的探究LINC01354调控脑胶质瘤细胞增殖、凋亡及焦亡的功能及机制。方法实时荧光定量聚合酶链反应(qRT-PCR)检测人小胶质细胞HMC3和脑胶质瘤细胞U251中LINC01354相对表达量。双荧光素酶报告基因实验验证LINC01354和miR-214-3p的靶向关系。人脑胶质瘤细胞U251分为空白组、sh-NC组、sh-LINC01354组、miR-214-3p mimic组、miR-NC组、sh-LINC01354+miR-214-3p inhibitor组和sh-LINC01354+inhibitor NC组。MTT法和流式细胞术检测细胞增殖和凋亡。Western blot检测细胞焦亡相关蛋白NOD样受体家族吡喃结构域包含蛋白3(NLRP3)、剪切的半胱天冬氨酸蛋白酶-1(Cleaved Caspase-1)和剪切的N-末端消皮素D(Cleaved N-terminal GSDMD)蛋白表达。结果人脑胶质瘤细胞U251中LINC01354表达高于人小胶质细胞HMC3(P<0.001),双荧光素酶报告基因结果显示miR-214-3p为LINC01354靶基因。MTT和流式细胞术检测结果显示,sh-LINC01354组U251细胞增殖能力低于sh-NC组(P<0.05),细胞凋亡高于sh-NC组(P<0.001),NLRP3、Cleaved Caspase-1和Cleaved N-terminal GSDMD蛋白表达均高于sh-NC组(P<0.01)。miR-214-3p mimic组U251细胞增殖能力均低于miR-NC组(P<0.05),细胞凋亡高于miR-NC组(P<0.001),NLRP3、Cleaved Caspase-1和Cleaved N-terminal GSDMD蛋白表达均高于miR-NC组(P<0.001)。sh-LINC01354+miR-214-3p inhibitor组U251细胞增殖高于sh-LINC01354+inhibitor NC组(P<0.05),细胞凋亡低于sh-LINC01354+inhibitor NC组(P<0.001)。结论LINC01354靶向抑制miR-214-3p影响U251细胞的增殖、凋亡及焦亡。 展开更多

胶质瘤是人类中枢神经系统最常见的恶性肿瘤,其早期诊断困难,复发率高,预后差[1]。胶质母细胞瘤(Glioblastoma Multiforme,GBM)是恶性和侵袭性最强的类型[2],在首次诊断后的中位生存时间为12-14个月[3]。由于胶质母细胞瘤增殖迅速、侵... 胶质瘤是人类中枢神经系统最常见的恶性肿瘤,其早期诊断困难,复发率高,预后差[1]。胶质母细胞瘤(Glioblastoma Multiforme,GBM)是恶性和侵袭性最强的类型[2],在首次诊断后的中位生存时间为12-14个月[3]。由于胶质母细胞瘤增殖迅速、侵袭性强且对化疗耐药,手术切除、放疗和化疗等方法的效果有限[4]。因此,新的治疗方法有待开发。在人类基因组中大约98%的转录本不编码蛋白质,这类RNA被称为非编码RNA(Non-coding RNA,ncRNA),其中长链非编码RNA(Long Noncoding RNA,lncRNA)约占ncRNA的80-90%,是由200多个核苷酸组成[5]。近年来,随着新一代测序技术的发展,越来越多的研究表明ncRNA在疾病发展中起着重要作用。 展开更多

目的:探讨细胞焦亡执行蛋白GSDMD在脑胶质瘤中的表达及其与患者预后之间的相关性。方法:本研究通过生物信息学方法分析TCGA和CGGA数据库中的脑胶质瘤和正常脑组织样本,对gasdermins(GSDMs)家族基因表达与患者预后进行分析筛选出相关性... 目的:探讨细胞焦亡执行蛋白GSDMD在脑胶质瘤中的表达及其与患者预后之间的相关性。方法:本研究通过生物信息学方法分析TCGA和CGGA数据库中的脑胶质瘤和正常脑组织样本,对gasdermins(GSDMs)家族基因表达与患者预后进行分析筛选出相关性最强的基因。回顾性收集2020年1月~2024年6月新疆医科大学第二附属医院神经外科收治的51例脑胶质瘤手术患者的临床资料及病理蜡块,按存活状态分为两组,对病理蜡块进行免疫组化实验测定GSDMD蛋白的表达水平。使用Kaplan-Meier生存分析和多因素Cox回归模型评估GSDMD表达与患者预后的关系。结果:GSDMD高表达是影响脑胶质瘤患者预后的独立危险因素(P<0.05);GSDMD高表达组的总生存时间显著短于低表达组(P<0.01);高表达GSDMD与脑胶质瘤高级别和较高的Ki-67指数相关(P<0.05)。结论:GSDMD高表达可能与脑胶质瘤患者的较差预后相关,并有潜力作为一个独立的预后不良指标。 展开更多

古陆相沉积型稀土矿是一种新型稀土矿,对稀土矿浸出渣进行金属元素分离提取,可提高稀土矿的利用价值和解决固废处置难题。本工作采用硫酸溶液浸出稀土矿浸出渣,探究硫酸溶液对Al,Fe和Ti浸出行为的影响和反应机理。研究结果表明,硫酸溶... 古陆相沉积型稀土矿是一种新型稀土矿,对稀土矿浸出渣进行金属元素分离提取,可提高稀土矿的利用价值和解决固废处置难题。本工作采用硫酸溶液浸出稀土矿浸出渣,探究硫酸溶液对Al,Fe和Ti浸出行为的影响和反应机理。研究结果表明,硫酸溶液可以有效溶解稀土矿中的硅铝酸盐和赤铁矿,选择性地浸出Al和Fe,锐钛矿不易与硫酸反应,因此Ti的浸出率较低。通过单因素和正交实验优化反应条件,在反应温度115℃、反应时间6 h、酸渣质量比2.1 g/g和液固比4 g/g的最佳反应条件下,Al,Fe和Ti浸出率分别达到86.44%,94.00%和7.14%。浸出液通过硫酸铝铵法可将Al和Fe有效分离,并制得Al_(2)O_(3)。 展开更多

Objective:Gastric cancer(GC)is a globally common cancer characterized by high incidence and mortality worldwide.Advances in the molecular understanding of GC provide promising targets for GC diagnosis and therapy.Long non-coding RNAs(lncRNAs)and their downstream regulators are regarded to be implicated in the progression of multiple types of malignancies.Studies have shown that the lncRNA small nucleolar RNA host gene 4(SNHG4)serves as a tumor promoter in various malignancies,while its function in GC has yet to be characterized.Therefore,our study aimed to explore the role and underlying mechanism of SNHG4 in GC.Methods:We used qRT-PCR to analyze SNHG4 expression in GC tissues and cells.Kaplan-Meier analysis was used to assess the correlation between SNHG4 expression and the survival rate of GC patients.Cellular function experiments such as CCK-8,BrdU,colony formation,flow cytometry analysis,and transwell were performed to explore the effects of SNHG4 on GC cell proliferation,apoptosis,cell cycle,migration,and invasion.We also established xenograft mouse models to explore the effect of SNHG4 on GC tumor growth.Mechanically,dual luciferase reporter assay was used to verify the interaction between SNHG4 and miR-409-3p and between miR-409-3p and cAMP responsive element binding protein 1(CREB1).Results:The results indicated that SNHG4 was overexpressed in GC tissues and cell lines,and was linked with poor survival rate of GC patients.SNHG4 promoted GC cell proliferation,migration,and invasion while inhibiting cell apoptosis and cell cycle arrest in vitro.The in vivo experiment indicated that SNHG4 facilitated GC tumor growth.Furthermore,SNHG4 was demonstrated to bind to miR-409-3p.Moreover,CREB1 was directly targeted by miR-409-3p.Rescue assays demonstrated that miR-409-3p deficiency reversed the suppressive impact of SNHG4 knockdown on GC cell malignancy.Additionally,miR-409-3p was also revealed to inhibit GC cell proliferation,migration,and invasion by targeting CREB1.Conclusion:In conclusion,we verified that the SNHG4 promoted GC growth and metastasis by binding to miR-409-3p to upregulate CREB1,which may deepen the understanding of the underlying mechanism in GC development. 展开更多

椎间盘突出症是骨科的高发疾病,而作为其关键病理基础的椎间盘退行性病变(intervertebral disc degeneration,IDD)是一个以细胞外基质进行性降解、结构破坏与生物力学功能丧失为特征的复杂病理过程,不仅在人群中呈现出更高的患病率,更... 椎间盘突出症是骨科的高发疾病,而作为其关键病理基础的椎间盘退行性病变(intervertebral disc degeneration,IDD)是一个以细胞外基质进行性降解、结构破坏与生物力学功能丧失为特征的复杂病理过程,不仅在人群中呈现出更高的患病率,更是导致全球人类慢性腰背痛与功能障碍的首要原因,造成了巨大的社会经济负担。尽管构建IDD动物模型对探索该疾病病理机制与推动转化研究具有重要意义,但目前关于IDD的病因与病理生理机制尚未完全阐明,且人类与常见实验动物在脊柱解剖、生物力学及退变病程上存在显著差异,加之现有的IDD动物模型繁杂多样且缺乏统一标准。因此,本指南系统梳理了啮齿类动物、非人灵长类动物以及兔、绵羊/山羊、猪、犬等不同动物的IDD模型,重点阐述三类主流模型的建模原理:诱发性模型(如纤维环/髓核/终板损伤和机械力学损伤)可控性强、周期短,适用于模拟急性损伤与快速筛选疗法;自发性模型能更好地模拟人类与年龄相关的自然退变进程;基因修饰模型则为解析特定分子通路提供了有力工具。指南深入剖析了这些模型的关键技术要点、可重复性与临床相关性,并比较其优势、局限及适用研究场景,旨在引导研究者进行“以科学问题为导向”的精准模型选择。同时,为提升研究结果的深度与可比性,本指南提出了涵盖影像学、组织学、生物化学与分子生物学、生物力学及疼痛行为学等多维度的IDD动物模型实验终点评估体系与推荐观察时间窗,并明确了贯穿实验全程的“替代、减少、优化(replacement,reduction,refinement,3Rs)”伦理原则与动物福利要求。此外,指南还展望了结合单细胞组学、多尺度力学分析及加强疼痛表型评估等未来研究方向。本指南旨在为研究者在具体科学问题与资源条件约束下,如何规范化选择与应用IDD动物模型提供一套系统化、标准化的方法学框架,以期减少研究间的异质性,提升临床前研究成果的转化效率,促进本领域的高质量发展,最终为开发延缓乃至逆转IDD的创新疗法提供坚实的科学基础。 展开更多

Objective:Adverse-risk acute myeloid leukemia(AML)patients should receive allogeneic hematopoietic stem cell transplantation(allo-HSCT)at first complete remission(CR1).However,the influence of prior therapies[i.e.,venetoclax plus azacitidine(VEN-AZA)or intensive chemotherapy(IC)]on post-transplant outcomes remains inconclusive.This multicenter,retrospective study compared the post-transplant outcomes between patients receiving VEN-AZA and those receiving IC before allo-HSCT.Methods:This study was based on the transplant database of TROPHY group.Consecutive adverse-risk AML patients receiving allo-HSCT from January 2021 to June 2023 were screened in five Chinese transplant centers.Patients were categorized into VEN-AZA group if they received venetoclax combined with azacitidine as first-line therapy followed by allo-HSCT.Patients who received first-line therapy consisting of a mainstay treatment of cytarabine and anthracycline followed by allo-HSCT were categorized into IC group.Results:In the total cohort,the 3-year probabilities of overall survival,leukemia-free survival,and event-free survival were better in the IC group than VEN-AZA group,particularly for patients with ASXL1 mutations or SF3B1 mutations.However,the survival of the VEN-AZA group was not superior to that of IC group in patients aged≥55 years or those with the hematopoietic cell transplantation-comorbidity index scores≥1 before allo-HSCT.After propensity score matching(median age:VEN-AZA group:57 years;IC group:55 years),only the probability of overall survival for the IC group was better than that of VEN-AZA group(93.6%vs.78.0%,P=0.034)at the 1-year follow-up;however,all of the other clinical outcomes were comparable between the VEN-AZA and IC groups.The TP53 mutation was independently associated with post-transplant relapse and survival.Conclusions:Our results suggest that IC remains the cornerstone of therapy,whereas VEN-AZA may also be used in younger patients and medically fit patients with adverse-risk AML who are receiving allo-HSCT in CR1. 展开更多

The prevalence of Class Ⅲ malocclusion varies among different countries and regions. The populations from Southeast Asian countries (Chinese and Malaysian) showed the highest prevalence rate of 15.8%, which can seriously affect oral function, facial appearance, and mental health. As anterior crossbite tends to worsen with growth, early orthodontic treatment can harness growth potential to normalize maxillofacial development or reduce skeletal malformation severity, thereby reducing the difficulty and shortening the treatment cycle of later-stage treatment. This is beneficial for the physical and mental growth of children. Therefore,early orthodontic treatment for Class Ⅲ malocclusion is particularly important. Determining the optimal timing for early orthodontic treatment requires a comprehensive assessment of clinical manifestations, dental age, and skeletal age, and can lead to better results with less effort. Currently, standardized treatment guidelines for early orthodontic treatment of Class Ⅲ malocclusion are lacking. This review provides a comprehensive summary of the etiology, clinical manifestations, classification, and early orthodontic techniques for Class Ⅲ malocclusion, along with systematic discussions on selecting early treatment plans. The purpose of this expert consensus is to standardize clinical practices and improve the treatment outcomes of Class Ⅲ malocclusion through early orthodontic treatment. 展开更多

The undeformed chip thickness and grinding force are key parameters for revealing the material removal mechanism in the grinding process.However,they are difficult to be well expressed due to the ununiformed protrusion height and random position distribution of abrasive grains on the abrasive wheel surface.This study investigated the distribution of undeformed chip thickness and grinding force considering the non-uniform characteristics of abrasive wheel in the grinding of K4002 nickel-based superalloy.First,a novel grinding force model was established through a kinematic-geometric analysis and a grain-workpiece contact analysis.Then,a series of grinding experiments were conducted for verifying the model.The results indicate that the distribution of undeformed chip thickness is highly consistent with the Gaussian distribution formula.The increase in the grinding depth mainly leads to an increase in the average value of Gaussian distribution.On the contrary,the increase in the workpiece infeed speed or the decrease in the grinding speed mainly increases the standard deviation of Gaussian distribution.The average and maximum errors of the grinding force model are 4.9%and 14.6%respectively,indicating that the model is of high predication accuracy. 展开更多

Copper and cobalt were recovered from SICOMINES mining waste rock in the Democratic Republic of Congo.The process mineralogy of the samples was analyzed using scanning electron microscopy and energy dispersive spectroscopy.The results showed that copper minerals exhibited various forms and uneven particle sizes,while cobalt existed in the form of highly dispersed asbolane,and large amounts of easily slimed gangue minerals were filled in the samples,making it difficult to separate copper and cobalt minerals.The particle size range plays a decisive role in selecting the separation method for the copper−cobalt ore.Gravity separation was suitable for particles ranging from 43 to 246μm,while flotation was more effective for particles below 43μm.After ore grinding and particle size classification,applying a combined gravity separation(shaking table)−flotation method yielded concentrated minerals with a copper recovery of 72.83%and a cobalt recovery of 31.13%. 展开更多

Lithium-ion batteries with LiCoO_(2)(LCO)cathodes are widely used in various electronic devices,resulting in a large amount of spent LCO(SLCO).Therefore,there is an urgent need for an efficient technique for recycling SLCO.However,due to the presence of cobalt oxide with a spinel phase on the surface of highly-degraded LCO,the strong electrostatic repulsion from the transition metal octahedron poses a high Li replenishment barrier,making the regeneration of highly-degraded LCO a challenge.Herein,we propose a structural transformation strategy for reconstructing Li replenishment channels to aid the direct regeneration of highly-degraded LCO.In this approach,ball milling is employed to disrupt the inherent structure of highly-degraded LCO,thereby releasing the internal stress and converting the surface spinel phase into a homogeneous amorphous structure,which promotes Li insertion and regeneration.The regenerated LCO(RLCO)exhibits an outstanding discharge capacity of 179.10 mAh·g^(−1) in the voltage range of 3.0–4.5 V at 0.5 C.The proposed strategy is an effective regeneration approach for highly-degraded LCO,thereby facilitating the efficient recycling of spent lithium-ion battery cathode materials. 展开更多

Protrusive facial deformities,characterized by the forward displacement of the teeth and/or jaws beyond the normal range,affect a considerable portion of the population.The manifestations and morphological mechanisms of protrusive facial deformities are complex and diverse,requiring orthodontists to possess a high level of theoretical knowledge and practical experience in the relevant orthodontic field.To further optimize the correction of protrusive facial deformities,this consensus proposes that the morphological mechanisms and diagnosis of protrusive facial deformities should be analyzed and judged from multiple dimensions and factors to accurately formulate treatment plans.It emphasizes the use of orthodontic strategies,including jaw growth modification,tooth extraction or non-extraction for anterior teeth retraction,and maxillofacial vertical control.These strategies aim to reduce anterior teeth and lip protrusion,increase chin prominence,harmonize nasolabial and chin-lip relationships,and improve the facial profile of patients with protrusive facial deformities.For severe skeletal protrusive facial deformities,orthodonticorthognathic combined treatment may be suggested.This consensus summarizes the theoretical knowledge and clinical experience of numerous renowned oral experts nationwide,offering reference strategies for the correction of protrusive facial deformities. 展开更多

研究了不同温度(1140、1160、1180和1200℃)和时间(2和4 h)热校形对Ni_(3)Al基高温合金组织与性能的影响,通过JmatPro热力学软件计算不同温度下合金平衡相析出含量,采用扫描电镜(SEM)分析不同热校形温度和保温时间对合金中一次γ′相溶... 研究了不同温度(1140、1160、1180和1200℃)和时间(2和4 h)热校形对Ni_(3)Al基高温合金组织与性能的影响,通过JmatPro热力学软件计算不同温度下合金平衡相析出含量,采用扫描电镜(SEM)分析不同热校形温度和保温时间对合金中一次γ′相溶解程度及二次γ′相析出程度的影响,进而分析其对合金高温拉伸和高温持久性能的影响规律。结果表明:随着热校形温度的升高,合金中的一次粗大γ′相逐渐溶解,并在冷却的过程中逐渐析出二次细小均匀的γ′相。随着热校形保温时间的增加,二次细小γ′相析出越来越多。二次细小γ′相的含量直接影响合金的1100℃拉伸性能和1100℃/60 MPa条件下的持久性能。建议合金的热校形工艺为1160~1200℃,保温4 h,此时合金的性能达到最佳。 展开更多
