Background:The use of third-generation different tyrosine kinase inhibitors(TKIs)is considered the most effective option for treating advanced non-small cell lung cancer(aNSCLC)with epidermal growth factor receptor(EG...Background:The use of third-generation different tyrosine kinase inhibitors(TKIs)is considered the most effective option for treating advanced non-small cell lung cancer(aNSCLC)with epidermal growth factor receptor(EGFR)mutations.However,there is limited information on the efficacy and safety of aumolertinib in patients remains these cases.Methods:The clinical records of patients receiving aumolertinib as first-line therapy across four hospitals in the Guangxi Zhuang Autonomous Region from April 2020 to December 2021 were retrospectively analyzed,using progression-free survival(PFS)as the primary endpoint and overall survival(OS)representing the secondary endpoint.Adverse events(AEs)were assessed using the Common Terminology Criteria for Adverse Events(CTCAE v5.0).Results:Approximately 47 patients with EGFR-Mutant aNSCLC were recruited,including 1 squamous cell carcinoma(SCC)patient,1 EGFR G719C mutated patient,1 EGFR S768 patient mutated,and 1 EGFR KDD mutated patient.The average follow-up duration was 48.1 months concluding in August 2024.The median PFS(mPFS)was 22.2 months(95%CI 17.6 to 26.7),while the median OS(mOS)was 39.7 months(95%CI 32.6 to 46.9).Patients with deletion of exon19 in EGFR(19del)showeda mPFS of 28.4 months,markedlylonger than those with the L858R point mutation(L858R),who had a mPFS of 15.2 months(p<0.036).Overall,22 patients(46.8%)had central nervous system(CNS)metastases at the=basal level.The mPFS for this cohort was 19.7 months.Rashes(17.0%),skin decrustation(4.2%),pruritus(4.2%),dental ulcers(4.2%),increased creatine kinase(2.1%),and musculoskeletal pains(2.1%)were the most prevalent AEs in this study.Grade 3 and higher AEs were observed at a rate of 4.2%.Conclusion:This study concluded that aumolertinib has considerable safety and efficacy for EGFR-mutant NSCLC in a first-line defense.展开更多
The intensifying global issues of freshwater scarcity and antibiotic contamination,especially in coastal environments,present interconnected threats to both ecosystems and public health.Addressing these issues demands...The intensifying global issues of freshwater scarcity and antibiotic contamination,especially in coastal environments,present interconnected threats to both ecosystems and public health.Addressing these issues demands innovative solutions that synergistically enhance energy efficiency,promote sustainability,and deliver multifunctional benefits.In this study,we present a solar-driven photothermalphotocatalytic synergistic platform(SPSP)constructed from a PF/Co_(3)O_(4)/CNTs@O-ANF composite(PCCO),engineered to achieve simultaneous seawater desalination and antibiotic degradation.The strategically designed 3D hierarchical architecture combines broadband solar absorption,interfacial hydrophobic regulation,and catalytic heterojunction engineering,enabling an elevated water evaporation rate of 1.75 kg·m^(-2)·h^(–1) and efficient degradation of over 98%of tetracycline(TC)across three operational cycles.Outdoor field tests confirmed the system’s operational robustness,producing 6.82 kg·m^(–2)·day^(–1) of purified water.Comprehensive water quality analyses further verified the removal of more than 99%of dissolved salts and organic contaminants,with the collected water exhibiting a neutral pH and complete absence of residual antibiotic activity.More importantly,the purified water facilitated robust growth of Brassica rapa,resulting in a 210% increase in biomass relative to plants irrigated with contaminated water,thereby demonstrating both ecological safety and agricultural applicability.Collectively,this SPSP technology represents a substantial advancement in sustainable water treatment,offering an integrated,energy-efficient solution for producing clean water and effectively remedying antibiotics.展开更多

Two^(87)Sr optical lattice clocks(OLCs)are being developed and operated at the National Institute of Metrology(NIM),located on different campuses that are 40km apart.In order to compare the optical frequencies of thes...Two^(87)Sr optical lattice clocks(OLCs)are being developed and operated at the National Institute of Metrology(NIM),located on different campuses that are 40km apart.In order to compare the optical frequencies of these two Sr OLCs,a 58-km noise canceled fiber link is built to transfer both a 1542-nm transfer laser and a microwave reference from Changping campus to Hepingli campus.Two commercial optical frequency combs(OFCs)with adapted single-branch 698/1542nm outputs coherently unite the two 698-nm clock lasers and the 1542-nm transfer laser.The fractional instability of the comparison yields 3.1×10^(−17)at 10000 s averaging time.The measured fractional frequency difference between these two Sr OLCs was evaluated to be 1.9(3.2)×10^(−17),which is within their claimed uncertainties.This result demonstrates the consistency of their frequencies when they serve as optical frequency standards.Our remote comparison demonstrates the feasibility of optical clock comparison through a long-distance fiber link and contributes to the progress of redefinition of the SI second.展开更多

However, because the spatial resolution of remote sensing precipitation data is still low, it is difficult to meet the needs of hydrological research, which restricts their application in drought and flood analysis, hydrological simulation, etc. In response to this problem, this paper takes the Beijing-Tianjin-Hebei region as the research area, downscaling the TRMM data and the GPM data space of the continuation plan, and increasing the spatial resolution of the data to 1 km. This study established rat models of traumatic...Xuefu Zhuyu decoction has been used for treating traumatic brain injury and improving post-traumatic dysfunction, but its mechanism of action needs further investigation. This study established rat models of traumatic brain injury by controlled cortical impact. Rat models were intragastrically administered 9 and 18 g/kg Xuefu Zhuyu decoction once a day for 14 or 21 days. Changes in neurological function were assessed by modified neurological severity scores and the Morris water maze. Immunohistochemistry, western blot assay, and re- verse-transcription polymerase chain reaction were used to analyze synapsin protein and mRNA expression at the injury site of rats. Our results showed that Xuefu Zhuyu decoction visibly improved neurological function of rats with traumatic brain injury. These changes were accompanied by increased expression of synaptophysin, synapsin I, and postsynaptic density protein-95 protein and mRNA in a dose-de- pendent manner. These findings indicate that Xuefu Zhuyu decoction increases synapsin expression and improves neurological deficits alder traumatic brain injury.展开更多
Objective:Recurrent ovarian cancer is often resistant to drugs such as paclitaxel.Short hairpin RNA(shRNA)targeting MDRI,a gene involved in the process of drug resistance,may be a promising strategy to overcome drug r...Objective:Recurrent ovarian cancer is often resistant to drugs such as paclitaxel.Short hairpin RNA(shRNA)targeting MDRI,a gene involved in the process of drug resistance,may be a promising strategy to overcome drug resistance.Methods:Construction and identification of eukaryotic expression plasmid was transiently transfected into human ovarian cancer ce plasmid of shRNA targeting on MDR1 gene.The ne A2780/Taxol.Apoptosis was determined by flow cytometry using annexin V-FITC/PI double labeling.Expression of MDRI mRNA was detected by quantitative polymerase chain reaction(qPCR)and P-glycoprotein expression was detected using Western blot.Results The IC50 of paclitaxel in MDR1 shRNA-transfected group was significantly reduced(1.986±0.153)μmol/ml as compared with that in negative control(5.246±0.107)μmol/ml and empty vector-transfected group(5.212±0.075)μmol/ml(P〈0.05).The percent of the relative reverse sensitivity to paclitaxel on A2780/Taxol cells was 67.1%,and the apoptotic rate was significantly increased[(6.977±0.333)%]compared with control[(1.637±0.111)%]and empty vector-transfected group[(1.663±0.114)%](P〈0.05).Expressions of MDR1 mRNA and P-glycoprotein were significantly reduced compared with control(P〈0.05).Conclusion:The present study demonstrated that the eukaryotic expression plasmid of shRNA targeting on MDRI inhibited the expression of MDRI effectively,thus enhance the sensitivity of A2780/Taxol cells to paclitaxel.展开更多
Background The alexithymia trait is of high clinical interest.The Perth Alexithymia Questionnaire(PAQ)was recently developed to enable detailed facet-level and valence-specific assessments of alexithymia.Aims In this ...Background The alexithymia trait is of high clinical interest.The Perth Alexithymia Questionnaire(PAQ)was recently developed to enable detailed facet-level and valence-specific assessments of alexithymia.Aims In this paper,we introduce the first Chinese version of the PAQ and examine its psychometric properties and clinical applications.Methods In Study 1,the PAQ was administered to 990 Chinese participants.We examined its factor structure,internal consistency,test-retest reliability,as well as convergent,concurrent and discriminant validity.In Study 2,four groups,including a major depressive disorder(MDD)group(n=50),a matched healthy control group for MDD(n=50),a subclinical depression group(n=50)and a matched healthy control group for subclinical depression(n=50),were recruited.Group comparisons were conducted to assess the clinical relevance of the PAQ.Results In Study 1,the intended five-factor structure of the PAQ was found to fit the data well.The PAQ showed good internal consistency and test-retest reliability,as well as good convergent,concurrent and discriminant validity.In Study 2,the PAQ was able to successfully distinguish the MDD group and the subclinical depression group from their matched healthy controls.Conclusions The Chinese version of the PAQ is a valid and reliable instrument for comprehensively assessing alexithymia in the general population and adults with clinical/subclinical depression.展开更多

Methods From January 2012 to January 2013, 15 patients underwent primary bilateral retromuscular breast augmentation with round textured silicone-gel implants and a novel infra-nipple broken line incision. Preoperatively, a semicircular incision was marked along the inferior base of the nipple. It was then extended bilaterally using two transverse right-angled geometric broken lines within the pigmented areolar skin. Follow-up was performed to evaluate the sensation of nipple-areolar complex, the scar, and the shape and texture of the breasts. Results The average follow-up was 6.7 months. Most of the patients complained of paresthesia of the nipple or breast skin, but transient decreased sensation improved within 3 months. No patients showed permanent sensory changes of the nipple areolar complex at a minimum follow-up of 4 months. The scars were imperceptible in all patients. Conclusion We believe that for selected patients, the infra-nipple broken line incision is a practical and reliable method to achieve aesthetic result.展开更多
In drilling a deepwater well,the mud density window is narrow,which needs a precise pressure control to drill the well to its designed depth.Therefore,an accurate characterization of annular flow between the drilling ...In drilling a deepwater well,the mud density window is narrow,which needs a precise pressure control to drill the well to its designed depth.Therefore,an accurate characterization of annular flow between the drilling riser and drilling string is critical in well control and drilling safety.Many other factors influencing the change of drilling pressure that should be but have not been studied sufficiently.We used numerical method to simulate the process of drill string rotation and vibration in the riser to show that the rotation and transverse vibration of drill string can increase the axial velocity in the annulus,which results in the improvement of the flow field in the annulus,and the effect on pressure loss and its fluctuation amplitude.In addition,there are also multiple secondary flow vortices in the riser annulus under certain eccentricity conditions,which is different from the phenomenon in an ordinary wellbore.The findings of this research are critical in safely controlling well drilling operation in the deepwater environment.展开更多
Ogura cytoplasmic male sterility(Ogura CMS)is extensively applied in hybrid seed production in cruciferous crops.However,the posttranscriptional molecular basis of Ogura CMS in cruciferous crops remains elusive.Here,a...Ogura cytoplasmic male sterility(Ogura CMS)is extensively applied in hybrid seed production in cruciferous crops.However,the posttranscriptional molecular basis of Ogura CMS in cruciferous crops remains elusive.Here,a data-independent acquisition-based proteomic approach coupled with a parallel reaction monitoring-based targeted proteomic assay was used to analyze the proteome dynamics of Ogura CMS cabbage line RM and its maintainer line RF during floral bud development to obtain insights into the mechanism underlying Ogura CMS in cruciferous crops.A total of 9162 proteins corresponding to 61464 peptides were identified in RM and RF floral buds.The proteomic fluctuation of RM was weaker than that of RF.Differences in protein expression between RM and RF gradually enlarged with floral bud development.Fifteen continually up-regulated and eight continually down-regulated proteins were found in RM relative to RF throughout floral bud development.Differentially expressed proteins between RM and RF during floral bud development were implicated in the endoplasmic reticulum(ER)-associated protein processing pathway,in which most of them exhibited down-regulated expression in RM.These data suggest that ER-associated protein processing may be involved in pollen abortion in Ogura CMS cabbage by inhibiting the expression of critical factors.Our findings not only deepen the understanding of the molecular mechanisms of Ogura CMS in cruciferous crops but also provide better guidance for applying Ogura CMS in the hybrid breeding of cruciferous crops.展开更多
Background:Nasopharyngeal carcinoma(NPC)exhibits a significant prevalence in the southern regions of China,and paclitaxel(PTX)is frequently employed as a medication for managing advanced NPC.However,drug resistance is...Background:Nasopharyngeal carcinoma(NPC)exhibits a significant prevalence in the southern regions of China,and paclitaxel(PTX)is frequently employed as a medication for managing advanced NPC.However,drug resistance is typically accompanied by a poor prognosis.Exploring the synergistic potential of combining multiple chemotherapeutic agents may represent a promising avenue for optimizing treatment efficacy.Methods:This study investigated whether 3-Methyladenine(3-MA)could potentiated the effect of PTX and its potential molecular mechanism.Samples were divided into the following categories:Negative control(NC)with the solvent dimethyl sulfoxide(DMSO,0.5%v/v),PTX(400 nM),3-MA(4 mM),and PTX(400 nM)+3-MA(4 mM).The viability of NPC cells was assessed using both the cell counting kit-8(CCK-8)assay and the colony formation assay.Microscopic observation was performed to identify morphological cell changes.Flow cytometry was used to assess cell cycle status,mitochondrial membrane potential(MMP),and apoptotic cells.Western blotting was conducted to quantify the protein expression.Results:3-MA enhanced PTX-specific inhibition of NPC cell proliferation.PTX,either alone or in combination with 3-MA,caused cell cycle halt at the G2/M phase in the majority of NPC cells,and the combination treatment of PTX with 3-MA induced a higher rate of NPC cell death compared to PTX alone.Western blotting results revealed the combination of PTX with 3-MA heightened activation of cyclin-dependent kinase 1(CDK1),a key molecule in shifting cells from mitotic arrest to apoptosis,led to a reduction in Myeloid Cell Leukemia 1(MCL-1)expression and an increase in Poly(ADP-ribose)polymerase(PARP)cleavage.Conclusion:The concurrent administration of PTX with 3-MA effectively enhances PTX’s inhibitory impact on NPC and activates the apoptosis signal regulated by CDK1.展开更多
Many studies have investigated how the online community has informed language acquisition and teaching.In the scholarship of Computer-Mediated Communication Theory(CMC),while most of the studies have emphasized second...Many studies have investigated how the online community has informed language acquisition and teaching.In the scholarship of Computer-Mediated Communication Theory(CMC),while most of the studies have emphasized second language English writing using the CMC framework,evidence is shown to support CMC in foreign language instruction.However,there is a lack of research on how feedback from native speakers of Chinese influences Chinese learners'writing in a web environment.In response to the research gap,drawing on sociocultural and CMC,the current exploratory study probes the nature of feedback on Zhihu,the largest Chinese social question-and-answer website,and how feedback affects advanced Chinese learners'writing performance.Eight American university students learning Chinese as second language participated by completing two writing tasks on Zhihu.After posting essays on the website,these participants read and responded to online feedback within two weeks,followed by a revision of their original posts.Tokens of feedback,revised language items and learners'response to feedback were coded by the researcher.Findings reported feedback types from Zhihu users,exchanges between learners and feedback providers,and correlations between first and second drafts.Feedback on Zhihu is proven to be beneficial to advanced Chinese learners'writing performance,and pedagogical implication is suggested for the inclusion of Zhihu in advanced Chinese curriculum.展开更多

文摘The implementation of the ecological protection red line serves as a crucial protective barrier for the conservation of wild plants with extremely small populations by delineating specific geographical areas designated for their conservation.The implementation of a comprehensive set of conservation measures aimed at establishing a biodiversity conservation network for wild plants with extremely small populations has laid the foundation for the health and stability of ecosystems,as well as the harmonious coexistence of humans and the natural environment.Research on the conservation of wild plants with extremely small populations serves as a critical foundation for informing and enhancing the effective conservation of such populations.This paper provides a systematic examination of the theoretical and practical conservation status of certain wild plants with extremely small populations and discusses the conservation trends of these populations within the framework of ecological protection red line delineation.In the future,strategies for the conservation of wild plants with extremely small populations should be integrated with an analysis of the causes of endangerment.This approach will provide a robust foundation for applied research focused on the conservation of these vulnerable plant populations.

文摘Two^(87)Sr optical lattice clocks(OLCs)are being developed and operated at the National Institute of Metrology(NIM),located on different campuses that are 40km apart.In order to compare the optical frequencies of these two Sr OLCs,a 58-km noise canceled fiber link is built to transfer both a 1542-nm transfer laser and a microwave reference from Changping campus to Hepingli campus.Two commercial optical frequency combs(OFCs)with adapted single-branch 698/1542nm outputs coherently unite the two 698-nm clock lasers and the 1542-nm transfer laser.The fractional instability of the comparison yields 3.1×10^(−17)at 10000 s averaging time.The measured fractional frequency difference between these two Sr OLCs was evaluated to be 1.9(3.2)×10^(−17),which is within their claimed uncertainties.This result demonstrates the consistency of their frequencies when they serve as optical frequency standards.Our remote comparison demonstrates the feasibility of optical clock comparison through a long-distance fiber link and contributes to the progress of redefinition of the SI second.
文摘<div style="text-align:justify;"> Precipitation is an important part of the global hydrological cycle. The large- scale, high-precision continuous precipitation data obtained by satellite remote sensing detection technology has become an important source of spatial precipitation data. However, because the spatial resolution of remote sensing precipitation data is still low, it is difficult to meet the needs of hydrological research, which restricts their application in drought and flood analysis, hydrological simulation, etc. In response to this problem, this paper takes the Beijing-Tianjin-Hebei region as the research area, downscaling the TRMM data and the GPM data space of the continuation plan, and increasing the spatial resolution of the data to 1 km. Compared with the original data, spatial downscaling data not only greatly improves the spatial resolution, but also increases the accuracy of the data, which has better applicability. </div>

文摘Xuefu Zhuyu decoction has been used for treating traumatic brain injury and improving post-traumatic dysfunction, but its mechanism of action needs further investigation. This study established rat models of traumatic brain injury by controlled cortical impact. Rat models were intragastrically administered 9 and 18 g/kg Xuefu Zhuyu decoction once a day for 14 or 21 days. Changes in neurological function were assessed by modified neurological severity scores and the Morris water maze. Immunohistochemistry, western blot assay, and re- verse-transcription polymerase chain reaction were used to analyze synapsin protein and mRNA expression at the injury site of rats. Our results showed that Xuefu Zhuyu decoction visibly improved neurological function of rats with traumatic brain injury. These changes were accompanied by increased expression of synaptophysin, synapsin I, and postsynaptic density protein-95 protein and mRNA in a dose-de- pendent manner. These findings indicate that Xuefu Zhuyu decoction increases synapsin expression and improves neurological deficits alder traumatic brain injury.

文摘Objective:Recurrent ovarian cancer is often resistant to drugs such as paclitaxel.Short hairpin RNA(shRNA)targeting MDRI,a gene involved in the process of drug resistance,may be a promising strategy to overcome drug resistance.Methods:Construction and identification of eukaryotic expression plasmid was transiently transfected into human ovarian cancer ce plasmid of shRNA targeting on MDR1 gene.The ne A2780/Taxol.Apoptosis was determined by flow cytometry using annexin V-FITC/PI double labeling.Expression of MDRI mRNA was detected by quantitative polymerase chain reaction(qPCR)and P-glycoprotein expression was detected using Western blot.Results The IC50 of paclitaxel in MDR1 shRNA-transfected group was significantly reduced(1.986±0.153)μmol/ml as compared with that in negative control(5.246±0.107)μmol/ml and empty vector-transfected group(5.212±0.075)μmol/ml(P〈0.05).The percent of the relative reverse sensitivity to paclitaxel on A2780/Taxol cells was 67.1%,and the apoptotic rate was significantly increased[(6.977±0.333)%]compared with control[(1.637±0.111)%]and empty vector-transfected group[(1.663±0.114)%](P〈0.05).Expressions of MDR1 mRNA and P-glycoprotein were significantly reduced compared with control(P〈0.05).Conclusion:The present study demonstrated that the eukaryotic expression plasmid of shRNA targeting on MDRI inhibited the expression of MDRI effectively,thus enhance the sensitivity of A2780/Taxol cells to paclitaxel.

文摘Background The alexithymia trait is of high clinical interest.The Perth Alexithymia Questionnaire(PAQ)was recently developed to enable detailed facet-level and valence-specific assessments of alexithymia.Aims In this paper,we introduce the first Chinese version of the PAQ and examine its psychometric properties and clinical applications.Methods In Study 1,the PAQ was administered to 990 Chinese participants.We examined its factor structure,internal consistency,test-retest reliability,as well as convergent,concurrent and discriminant validity.In Study 2,four groups,including a major depressive disorder(MDD)group(n=50),a matched healthy control group for MDD(n=50),a subclinical depression group(n=50)and a matched healthy control group for subclinical depression(n=50),were recruited.Group comparisons were conducted to assess the clinical relevance of the PAQ.Results In Study 1,the intended five-factor structure of the PAQ was found to fit the data well.The PAQ showed good internal consistency and test-retest reliability,as well as good convergent,concurrent and discriminant validity.In Study 2,the PAQ was able to successfully distinguish the MDD group and the subclinical depression group from their matched healthy controls.Conclusions The Chinese version of the PAQ is a valid and reliable instrument for comprehensively assessing alexithymia in the general population and adults with clinical/subclinical depression.
文摘Objective To investigate reliability of the infra-nipple broken line incision for breast augmentation. Methods From January 2012 to January 2013, 15 patients underwent primary bilateral retromuscular breast augmentation with round textured silicone-gel implants and a novel infra-nipple broken line incision. Preoperatively, a semicircular incision was marked along the inferior base of the nipple. It was then extended bilaterally using two transverse right-angled geometric broken lines within the pigmented areolar skin. Follow-up was performed to evaluate the sensation of nipple-areolar complex, the scar, and the shape and texture of the breasts. Results The average follow-up was 6.7 months. Most of the patients complained of paresthesia of the nipple or breast skin, but transient decreased sensation improved within 3 months. No patients showed permanent sensory changes of the nipple areolar complex at a minimum follow-up of 4 months. The scars were imperceptible in all patients. Conclusion We believe that for selected patients, the infra-nipple broken line incision is a practical and reliable method to achieve aesthetic result.

文摘In drilling a deepwater well,the mud density window is narrow,which needs a precise pressure control to drill the well to its designed depth.Therefore,an accurate characterization of annular flow between the drilling riser and drilling string is critical in well control and drilling safety.Many other factors influencing the change of drilling pressure that should be but have not been studied sufficiently.We used numerical method to simulate the process of drill string rotation and vibration in the riser to show that the rotation and transverse vibration of drill string can increase the axial velocity in the annulus,which results in the improvement of the flow field in the annulus,and the effect on pressure loss and its fluctuation amplitude.In addition,there are also multiple secondary flow vortices in the riser annulus under certain eccentricity conditions,which is different from the phenomenon in an ordinary wellbore.The findings of this research are critical in safely controlling well drilling operation in the deepwater environment.

文摘Ogura cytoplasmic male sterility(Ogura CMS)is extensively applied in hybrid seed production in cruciferous crops.However,the posttranscriptional molecular basis of Ogura CMS in cruciferous crops remains elusive.Here,a data-independent acquisition-based proteomic approach coupled with a parallel reaction monitoring-based targeted proteomic assay was used to analyze the proteome dynamics of Ogura CMS cabbage line RM and its maintainer line RF during floral bud development to obtain insights into the mechanism underlying Ogura CMS in cruciferous crops.A total of 9162 proteins corresponding to 61464 peptides were identified in RM and RF floral buds.The proteomic fluctuation of RM was weaker than that of RF.Differences in protein expression between RM and RF gradually enlarged with floral bud development.Fifteen continually up-regulated and eight continually down-regulated proteins were found in RM relative to RF throughout floral bud development.Differentially expressed proteins between RM and RF during floral bud development were implicated in the endoplasmic reticulum(ER)-associated protein processing pathway,in which most of them exhibited down-regulated expression in RM.These data suggest that ER-associated protein processing may be involved in pollen abortion in Ogura CMS cabbage by inhibiting the expression of critical factors.Our findings not only deepen the understanding of the molecular mechanisms of Ogura CMS in cruciferous crops but also provide better guidance for applying Ogura CMS in the hybrid breeding of cruciferous crops.

文摘Background:Nasopharyngeal carcinoma(NPC)exhibits a significant prevalence in the southern regions of China,and paclitaxel(PTX)is frequently employed as a medication for managing advanced NPC.However,drug resistance is typically accompanied by a poor prognosis.Exploring the synergistic potential of combining multiple chemotherapeutic agents may represent a promising avenue for optimizing treatment efficacy.Methods:This study investigated whether 3-Methyladenine(3-MA)could potentiated the effect of PTX and its potential molecular mechanism.Samples were divided into the following categories:Negative control(NC)with the solvent dimethyl sulfoxide(DMSO,0.5%v/v),PTX(400 nM),3-MA(4 mM),and PTX(400 nM)+3-MA(4 mM).The viability of NPC cells was assessed using both the cell counting kit-8(CCK-8)assay and the colony formation assay.Microscopic observation was performed to identify morphological cell changes.Flow cytometry was used to assess cell cycle status,mitochondrial membrane potential(MMP),and apoptotic cells.Western blotting was conducted to quantify the protein expression.Results:3-MA enhanced PTX-specific inhibition of NPC cell proliferation.PTX,either alone or in combination with 3-MA,caused cell cycle halt at the G2/M phase in the majority of NPC cells,and the combination treatment of PTX with 3-MA induced a higher rate of NPC cell death compared to PTX alone.Western blotting results revealed the combination of PTX with 3-MA heightened activation of cyclin-dependent kinase 1(CDK1),a key molecule in shifting cells from mitotic arrest to apoptosis,led to a reduction in Myeloid Cell Leukemia 1(MCL-1)expression and an increase in Poly(ADP-ribose)polymerase(PARP)cleavage.Conclusion:The concurrent administration of PTX with 3-MA effectively enhances PTX’s inhibitory impact on NPC and activates the apoptosis signal regulated by CDK1.
文摘Many studies have investigated how the online community has informed language acquisition and teaching.In the scholarship of Computer-Mediated Communication Theory(CMC),while most of the studies have emphasized second language English writing using the CMC framework,evidence is shown to support CMC in foreign language instruction.However,there is a lack of research on how feedback from native speakers of Chinese influences Chinese learners'writing in a web environment.In response to the research gap,drawing on sociocultural and CMC,the current exploratory study probes the nature of feedback on Zhihu,the largest Chinese social question-and-answer website,and how feedback affects advanced Chinese learners'writing performance.Eight American university students learning Chinese as second language participated by completing two writing tasks on Zhihu.After posting essays on the website,these participants read and responded to online feedback within two weeks,followed by a revision of their original posts.Tokens of feedback,revised language items and learners'response to feedback were coded by the researcher.Findings reported feedback types from Zhihu users,exchanges between learners and feedback providers,and correlations between first and second drafts.Feedback on Zhihu is proven to be beneficial to advanced Chinese learners'writing performance,and pedagogical implication is suggested for the inclusion of Zhihu in advanced Chinese curriculum.
