银杏叶提取物对衰老大鼠急性肺损伤和肾功能受损的保护作用 被引量:11

Protective effect of Ginkgo biloba extract on acute lung injury in aging rats and renal function damage induced by acute lung injury

摘要 目的 观察银杏叶提取物 (GBE)对脂多糖 (LPS)诱发的衰老大鼠急性肺损伤及由此而诱导的肾功能受损是否有保护作用。 方法 雄性Wistar大鼠 36只复制成衰老模型。再随机分成对照组 (静脉注射生理盐水 ) ;LPS组 (静脉注射LPS)及GBE +LPS组。注LPS或盐水后 2h或 6h ,收集血 ,并制备肺、肾组织匀浆待测。 结果 衰老大鼠在注LPS后 2h、6h时已形成急性肺损伤。注LPS后 2h血中肌酐及尿素氮含量无显著性升高 ,至 6h时均显著升高 ,分别由对照组的 (6 8 7±6 9) μmol/L及 (5 9± 0 6 )mmol/L升至 (94 7± 10 3) μmol/L及 (11 4± 1 9)mmol/L (均为P <0 0 1)。注LPS后 2h ,血和肺组织中丙二醛和NO2 -/NO3 -含量均显著升高 (均为P <0 0 1) ;而此时 ,肺组织中谷胱甘肽过氧化物酶及Na+ K+ ATPase活性均显著下降 (均为P <0 0 1)。上述指标的变化维持至 6h时。而注LPS后 2h时 ,肾组织中丙二醛及NO2 -/NO3 -含量和谷胱甘肽过氧化物酶及Na+ K+ ATPase活性均无显著性改变。但至 6h时 ,肾组织中丙二醛和NO2 -/NO3 -含量均显著升高 (均为P <0 0 1)。而谷胱甘肽过氧化物酶和Na+ K+ ATPase活性均显著下降 (均为P <0 0 1)。事先给予GBE可显著地缓解血、肺及肾组织中上述指标的变化 (P <0 0 5 )。 Objective To investigate whether gingko biloba extract (GBE) have protective effect on acute lung injury(ALI)induced by Lipopolysaccharide ( LPS) in aging rats and renal function damage induced by ALI. Methods Male aging Wistar rats(36) were used in the study. LPS group (LPS, iv route, 5mg/kg body weight) and GBE+LPS group (GBE was given starting from 7 days before experiment, once a day oral administration). The blood, lung and kidney samples were collected at 2 or 6 hours after LPS or saline administration. Results Compared with the aging control, ALI was obviously observed in LPS group, blood creatinine and urea nitrogen were increased significantly from (68.7± 6.9) mol/L and (5.9±0.6) mmol/L to (94.7±10.3) mol/L and (11.4±1.9) mmol/L, respectively (P<0.01) at 6 hours after LPS administration. In LPS group, MDA and NO 2 -/NO 3 -contents in blood and lung tissue were all increased significantly (P<0.01) at 2 and 6 hours. But GSH-P X and Na +-K +-ATPase activities in lung tissue were decreased significantly (P<0.01) at 2 and 6 hours after LPS administration. Whereas MDA, NO 2 -/NO 3 - contents in renal tissue were increased obviously (P<0.01), and Na +-K +-ATPase and GSH-P X activities were decreased markedly (P<0.01) only at 6 hours after LPS administration. All these changes were significantly attenuated in the GBE+LPS group (P<0.05). Conclusions ALI was caused by intravenous injection of LPS (5 mg/kg body weight ) in aging rats. Renal function damage could be induced by ALI in aging rats. GBE showed a protective effect on ALI and renal function damage in this animal model.
