槲皮素对内毒素急性肺损伤的保护作用 被引量:24

Protective effects of quercetin in rats with lipopolysaccharide-induced acute lung injury

摘要 目的 探讨槲皮素 (quercetin)对内毒素急性肺损伤 (ALI)的保护作用。方法  2 4只SD大鼠 ,随机分成ALI模型组、槲皮素干预组、正常对照组 (n =8只 )。各组大鼠均于注射LPS或生理盐水 6h后测定PaO2 、pH值、PaCO2 ,血清TNF α、IL 8、P 选择素浓度和肺组织匀浆髓过氧化物酶 (MPO)含量及TNF α、IL 8、P 选择素浓度 ;观察肺病理改变、肺湿 /干重比 (W /D)。结果 与ALI模型组比较 ,槲皮素干预组的肺水肿病理改变明显减轻 ,PaO2 改善 ,MPO、P 选择素及TNF α、IL 8降低 (P <0 0 1)。结论 槲皮素能改善ALI大鼠的气体交换功能 ,抑制炎性介质的释放 。 Objective To investigate the protective effects of quercet in rats with lipopolysaccharide-induced acute lung injury(ALI).Methods Lipoplysaccharidel(LPS,6 mg/kg,iv)reproduced the model of ALI in rats.24 SD rats were randomly divided into three groups(n=8)?Six hours after lipopolysaccharide or normal saline was administered by intravenous injection,the levels of tumour necrosis factor alpha(TNF-α),interleukin-8(IL-8)and P-selectin in serum were measured by radioimmunoassay and arterial blood gases were analysed.The rats were sacrificed by bleeding at the end of experiment,the pathological changes of rat lungs were observed,the wet and dry weight ratio(W/D)was calculated,the concentration of lung myeloperoxidase(MPO)was detected by the method of colorimetry.The levels of plasma TNF-α,IL-8 and P-selectin of lung tissue were measured by radioimmunoassay.Results In ALI model group,the levels of W/D,PaCO 2,MPO,TNF-α,IL-8,P-selectin were increased significantly(P<0.01),whereas PaCO 2 and arterial pH were obviously decreased compared with control group(P<0.01).Compared with ALI model group these changes were markedly attenuted in Quercetin pretreatment group (P<0.05)and the pathological changes of the lung were also ameliorated.Conclusion Quercetin has a protective effect on LPS-induced acute lung injury in rats through inhibiting the inflammatory mediators,thus improving gas exchange ability.
