摘要
本文研究了白介素2(IL-2)和PHA诱导的LAK细胞对混杂于骨髓细胞中的U937、HL60和K562细胞的作用。将正常骨髓单个核细胞与白血病细胞按10:1比例混合,加入一定比例LAK细胞后孵育4、12小时,然后做白血病集落培养。结果显示,当LAK细胞:骨髓单个核细胞:白血病细胞为20:10:1,预孵育12小时,U937、HL60和K562集落生成分别为对照组之1.21%±2.10%,1.56%±1.65%和0.26%±0.19%。而相同条件下,骨髓CFU—c生长为对照组之87.0%±17.1%。表明在上述条件下,LAK细胞对骨髓中白血病细胞具有选择性杀伤/抑制效应。
The effect of interleukin-2 (IL-2) and phytohemaglutinin A (PHA) induced lymphokine-activated killer (LAK) cells on U937, HL60 and K562 cell lines in bone marrow was studied. Bone marrow mononuclear cells (BMMNC) were mixed with leukemic cells at the ratio of 10 :1, and LAK cells were added at certain concentration. Leukemia colony formation was assayed after preincubation of 4 and 12 hours. The data Showed that when LAK: BMMNC: leukemic cell was 20 : 10 : 1, the colony formation of U937, HL60 and K562 were 1.21% ±2.10% , 1.56% ±1.65% and 0.26 ±0.19% of control resectively after 12 hours preincubation. At the same condition, the number of CFU-c was 87.0% ±17.1% of control, indicating that LAK cells have selective cytotoxic/inhibitory effects on leukemic cells in bone marrow.
