摘要
应用Ames试验对碳素粉尘致突变检测前处理方法进行研究。比较索氏萃取法(Sexheltextraction及超声振动萃取法和不同萃取溶剂-环已烷、二氯甲烷及甲醇的萃取效果。经统计处理,选出萃取方法与溶剂的最佳搭配的前处理方法,移码突变检测-TA98(±S9),选用索氏萃取法、二氯甲烷溶剂。碱基置换致突变检测-TA100(±S9)选用超声振动萃取法、二氯甲烷溶剂。
The method of prchandling for assay of mutagcnicity of carbon dust by Ames test was studies. Two extractive methods (Soxihelt extraction and Ultrasonic vibratory extraction) are compared. The extractive effect of three solvents (Dichloromethane, Methanol and Cyclohexane) arc also compared. The test data are handled with variance of statstical analysis.The method of Soxhelt extraction using Dichloromethane as solvent is suggested to be chosen for framcshift mutagcns detected with TA98(± S9), while the method of ultrasonic vibration using Dichloromethane as solvent is chosen for base-pair substitution detected with TA100(± S9), while the method of ultrasonic vibration using Dich loromethane as solvent is chosen for base-pair substitution detected with TA100(± S9).
