摘要
研究化湿利水泄浊法治疗高血压的机理 ,组织贴块法行自发性高血压大鼠 (SHR)主动脉VSMC的分离培养 ,并以倒置显微镜、α SMactin免疫组化法鉴定 ;以MTT法观察含药血清对VSMC增殖的影响、3H TdR掺入法观察含药血清对VSMCDNA合成的影响 ,并与平肝潜阳法、西药依那普利比较。结果 :化湿利水泄浊法各浓度组对VSMC增殖均有抑制作用 ,且在 1 2 5 %~ 15 %之间有一定的量效关系 ;化湿利水泄浊法对VSMC的增殖抑制作用与依那普利相似优于平肝潜阳组 ;化湿利水泄浊法对VSMCDNA合成的抑制作用 ,优于对照组。结论 :化湿利水泄浊法对培养SHR的VSMC增殖有明显抑制作用 ,由此改善高血压血管重构。
To study the mechanism of dampness-transforming,water-disinhibiting and turbidity-resolving therapy for hypertension,vascular smooth muscle cells (VSMC) were isolated from the media of spontaneous hypertensive rats (SHR) aorta obtained by explant method and identified by inverted microscope and α-SM actin immunohistochemical method.The effects of the drug containing serum on cell proliferation were observed by MTT method,and the change of VSMC DNA synthesis by 3H-TdR incorporation method.And Pinggan Qianyang method and Enalaprol were used as control.Results:This therapy had inhibiting effects on the VSMC proliferation,with the concentration between 1.25%~15%;this therapy had similar effects to Enalaprol,but better effects than Pinggan Qianyang method;and this therapy could inhibit the VSMC DNA synthesis,with better effects than control group.It is indicated that this therapy has marked inhibitory effects on the proliferation of VSMC,and hence improve the vascular remodeling.
