摘要
目的构建TRP-1反义核酸、转染黑素细胞并观察细胞生长和黑素代谢功能的改变,探讨TRP-1在黑素细胞中的作用。方法将TRP-1反义核酸真核表达载体转染黑素细胞,以测定的MTT结果绘制细胞生长曲线;测定TRP-1反义核酸转染黑素细胞后细胞内黑素含量的变化。结果从生长曲线可以看出,TRP-1反义核酸转染的黑素细胞增殖速度明显低于对照细胞;黑素含量明显降低。结论TRP-1反义核酸能显著抑制黑素细胞生长并降低黑素含量,表明TRP-1在维持黑素细胞正常的生长和黑素代谢中发挥重要作用。
Objective To construct the antisense eukaryotic vector of human TRP-1 (tyrosinase related protein 1,TRP-1)encoding gene, and transfect its antisense vector into melanocytes to further study its effect on proliferation and function of melanocytes. Methods Antisense TRP-1 recombinant vector was transfected into melanocytes.According to the results of MTT,cell growth curves were drawn. Then, the content of melanin in antisense TRP-1 transfected melanocyte was detected. Results It could be seen from the cell growth curves, antisense TRP-1 transfected melanocytes grew at a relative low rate compared with the controls.In addition, the content of melanin in antisense TRP-1 transfected melanocytes decreased significantly. Conclusion Antisense TRP-1 could notably block the proliferation of melanocytes and decrease the content of melanin,which suggested that TRP-1 play an important role in the proliferation and melanin metabolism in melanocytes.
