摘要
采用生物素—抗生物素蛋白DCS体系间接免疫荧光新技术,以5种单克隆抗体研究22~28胚期鸡胚部分面部原基的细胞外间质(extracellular matrix,ECM)大分子的存在和分布。结果表明,ECM对胚胎器官形态发生中细胞的运动、粘附和锚定起了相当重要的作用,迅速向内生长的鼻基板基膜处的强荧光提示ECM对鸡胚鼻孔形成有诱导和调控作用。
The presence and distribution of extracellular matrix (ECM) macronole-cules were analyzed in the facial prinordia of chick embryos from stages (22-28) of development corresponding to naris fornation. Frozen sections through the nasal placode, medial nasal processes and lateral nasal processes were prepared for indirect immunofluorescence employing a biotin-avidin DCS system using monoclonal antibodies against type Ⅳ collagen, laminin and fibronectin. The results demonstrated that ECM played a very important role in regulating migration, adhesion and anchorage of cells during morphogenesis. Strong fluorescence of basal lamina was present in the areas of rapid ingrowth of the nasal placode. It was suggested that ECM has very important guiding and regulating role during naris formation in chick embryo.
