麻黄汤及其减桂枝对哮喘小鼠影响的比较研究 被引量:13

摘要 目的 :探讨麻黄汤及其减桂枝对哮喘小鼠的影响 ,了解桂枝在本方中的作用。方法 :4 8只小鼠随机分为对照组、哮喘组、麻黄汤治疗组、麻黄汤减桂枝治疗组。用卵蛋白致敏建立哮喘模型 ,观察哮喘小鼠肺组织病理变化及 γ 干扰素 (IFNγ)和白细胞介素 4 (IL 4 )在肺组织的表达情况。结果 :麻黄汤治疗组和麻黄汤减桂枝治疗组小鼠支气管内及其管壁浸润细胞数比哮喘组明显减少 (P均 <0 .0 1) ;麻黄汤治疗组支气管内及其管壁浸润细胞数的减少比麻黄汤减桂枝治疗组更为明显 (P<0 .0 5 )。麻黄汤治疗组和麻黄汤减桂枝治疗组肺组织 IFNγ阳性细胞数比哮喘组均明显增多 (P均 <0 .0 1) ,麻黄汤治疗组和麻黄汤减桂枝治疗组肺组织IL 4阳性细胞数比哮喘组均明显减少 (P均 <0 .0 1) ;但麻黄汤治疗组与麻黄汤减桂枝治疗组间肺组织 IFNγ和 IL 4阳性细胞数均无明显差别 (P均 >0 .0 5 )。结论 :麻黄汤、麻黄汤减桂枝均能增加哮喘小鼠肺组织IFNγ,降低其 IL 4的表达 ,这可能是其治疗哮喘的作用机制之一。麻黄汤减少哮喘小鼠支气管及其周围组织炎症细胞浸润的作用比麻黄汤减桂枝明显 。 Objective: To investigate the effects of Mahuang decoction(麻黄汤) and Mahuang decoction taken away Guizhi(桂枝) on asthmatic mouse, and observe the effects of Guizhi in Mahuang decoction on asthmatic mouse. Methods: Fortyeight mice were divided into control group, asthmatic group, Mahuang decoction treatment group and Mahuang decoction taken away Guizhi treated group. Asthmatic mouse model was established by intraperitoneal injection ovalbumin. After 3 weeks, and the asthma consecutively challenged daily with spray inhalation of 5% ovalbumin for 7 days. The pathologic change and the lung tissues expression of interleukin4(IL4) and γinterferon(IFNγ) of all groups mice were analyzed. Results: The number of inflammatory cell in or around the bronchi of the mouse in the two treatment groups were less than that in asthma group (both P<0 01), and the number of inflammatory cell in treated group with Mahuang decoction was less than that in treated group with Mahuang decoction taken away Guizhi (P<0 05). The expressions of IFNγ in the lung tissues of the mouse in the two treatment groups were more than that in asthmatic group(P<0 01). While the expressions of IL4 in the lung tissues of the mouse in the two treatment groups were less than that in asthmatic group(P<0 01). The expressions of IFNγ and IL4 in the lung tissues of the mouse in Mahuang decoction taken away Guizhi treatment group and Mahuang decoction treatment group were not different (both P>0 05). Conclusion: Mahuang decoction and Mahuang decoction taken away Guizhi can decrease IL4 and increase IFNγ, it's the mechanism of them in treating asthmatic mouse. Guizhi in Mahuang decoction can alleviate inflammation of bronchus of asthmatic mouse.
