摘要
目的 :探讨中药止哮平喘方对哮喘豚鼠模型 Th1/ Th2细胞的影响。方法 :采用免疫组织化学 (组化 )方法测定模型动物外周单个核细胞 (PBMC)中 CD+ 4、CD+ 8T细胞的数量 ;用原位末端脱氧核苷酸转移酶标记法(TU NEL )标记的流式细胞术检测 T淋巴细胞凋亡情况。结果 :哮喘模型组 CD+ 4T细胞高于正常对照组(P<0 .0 5 ) ;中药大剂量组 CD+ 4T细胞显著降低 (P<0 .0 5 )、CD+ 8T细胞与正常对照组无明显差异 (P>0 .0 5 )。哮喘模型组 T淋巴细胞凋亡率显著低于正常对照组 (P<0 .0 1) ;中药大剂量组 T淋巴细胞凋亡率显著增加(P<0 .0 5 )。结论 :中药止哮平喘方可降低 CD+ 4细胞数量 ,具有抗气道炎症作用 ,其诱导 T淋巴细胞凋亡可能是主要机制之一。
Objective: To investigate the effects of traditional Chinese herbs Zhixiao Pingchuan prescription(止哮平喘方) on Th1/Th2 cells in guineapigs asthma model. Methods: CD + 4 and CD + 8 T cells counts were detected with immunocytochemistry (ICC) in the peripheral blood mononuclear cells (PBMC).Apoptosis of T lymphocyte was detected by TdTmediated dUTP nick end labeling(TUNEL) method and flow cytometry assay . Results: The number of CD + 4 cells (Th2) was significantly increased in asthma group than that in the normal group ( P <0.05),and that of the highdose group was significantly decreased( P <0.05). There was no remarkable difference in the number of CD + 8 cells (Th1) compared to the normal group ( P >0.05). The percentages of apoptosis cells in T lymphocytes were significantly decreased in the asthma group than that in the normal group( P <0.01) .The percentages of apoptosis cells in T lymphocytes were significantly increased in the highdose group( P <0.05). Conclusion: Traditional Chinese herbs Zhixiao Pingchuan prescription can improve the airway inflammation, lowering the numbers of Th2 type cells (CD + 4 cells) by enhancing apoptosis of T lymphocyte.
