摘要
普通烟草(Nicotina tabacum L.c.v.Samsun)营养生长植株和开花植株表皮薄层在生长素和细胞分裂素配比下,可诱导花原基和成花。不同部位茎表皮薄层外植体的生化分析表明;α-淀粉酶活性向顶递增,而过氧化物酶活性及其同工酶水平向顶递减。在营养生长植株中内源游离 IAA 含量变化不显著,而在开花植株中,花梗表皮薄层游离 IAA 含量为主茎的两倍。这些生化梯度与外植体的体外花芽分化能力相联系。
In vitro differentiation of floral buds can be obtained from thin cell layer culture(oneto six layers of epidermal and subepidermal cells)of flower stalk sections of N.tabacum L.cv.Samsun by varying the auxin-cytokinin ratio in MS medium and cultural condition.Some biochemical analyses were conducted in relation to a“floral gradient”present inthin cell layers of vegetative and flowering plants.There were qualitative and quantitativedifferences in the isoperoxidases,with decrease in content and fewer bands acropetally,butthe α-amylase activities increased with the explant going from the base of the stem to theapex.The contents of free indole-3-acetic acid(IAA)did not show remarkable diffe-rence in stem sections of vegetative plants,but the IAA content in the explants of floralstalk was twice higher than those in the explants of main stem.These biochemical gra-dients were in connection with the floral bud differentiation of explants.
