摘要
采用猴 8只,单侧颈内动脉注射 1甲基-4苯基-1,2,3,6-四氢吡啶(MPTP),两周内出现一侧肢体的巴金森氏病症状,能诱发出旋转行为,服美多巴后可纠正旋转动作。肌电图检查;以震颤为主要症状者呈群放电位,以强直为主要表现者呈猝发高频电位。病理切片中,以震颤为主要症状的动物,中脑及丘脑下部的黑质细胞减少或消失。主要症状为强直者,除黑质病变外,基底神经节、兰斑及小脑浦肯野氏细胞均有减少或变性。实验表明该模型简便、可靠、重复性好。
In this paper, the 1-methyl-4-(2-methyl phenyl)-1, 2, 3. 6-tetrahydropyridine-induced parkinsonismmodel in monkey is reported and the pathologicalchanges are studied. After unilateral injection ofMPTP into internal carotid artery, the monkeyspresented parkinson-like symptoms on contralateralside within 2 weeks. Rotational behavior can befired by apomorphine and attenuated by madopa.The EMG of these experimental animals dividedinto athetosic movement and mascular rigiditytype. In the former type it was found the neuronsdecreased in substantia nigra. The later type wasmore complex. It was found cell degenerationand slight demyelination in basal ganglia andocus coeruleus. Purkinje's cells were also degenerated.The results indicated that a reliable, con-venient and reproducible model was produced forthe study of Parkinson disease and brain tissuetransplantation.
