二烯丙基二硫对人白血病细胞株增殖的影响

摘要 目的 研究二烯丙基二硫 (DADS)对人白血病细胞HL - 60的作用并探讨其作用机理。方法 采用MTT法、生长曲线绘制、流式细胞仪、免疫组化等方法 ,观察DADS对体外培养的HL - 60细胞的生长抑制、细胞周期分布以及PCNA表达的影响。结果 DADS对HL - 60细胞具有明显的生长抑制效应且呈剂量 -效应依赖关系 ,当 2 0mg·L- 1 DADS作用HL - 60细胞 72h ,增殖抑制率达 70 .5 %。流式细胞仪检测结果提示 :DADS处理后的HL - 60细胞主要停滞于S +G2 /M期 ,同时出现亚G1 峰 ;免疫组化结果显示 :DADS处理后的HL - 60细胞PCNA表达下降。结论 DADS对体外培养的HL - 60细胞具有明显的增殖抑制作用 ,DADS抑制HL - 60细胞增殖与细胞周期S +G2 Objective To explore the potential therapeutic effect of DADS on human leukemia cell line HL-60. Methods MTT was used to test the cell growth of HL-60, cell cycle and apoptosis of human leukemia cell line HL-60 were detected by flowcytometry (FCM), immunohistochemical staining detected the expression of PCNA. Results DADS significantly inhibited proliferation of HL-60 cell and the inhibiting effects had a dose-and time-dependence.The inhibition fraction of HL-60 cell exposed to 20 mg·L -1 DADS for 72 h was 70.5%, flow cytometry analysis revealed that HL-60 cell treated with DADS significantly increased in the percentage of cells in the G 2/M and S phase, and appearance of sub-G 1 peak in a dose-dependent manner. Immunohistochemical staining revealed that the expression of PCNA HL-60 cell reduced(P<0.05),when treated with 10 mg·L -1 DADS.Conclusion DADS can effectively inhibit the proliferation of human leukemia cell line HL-60 cell and the possible mechanisms may be related to G 2/M and S phase arrest and apoptosis induced by DADS.
