摘要
在稳定的普鲁士蓝修饰电极上先涂一酶层 ,然后将二氧化硅溶胶 -凝胶薄膜敷着其上用于固定酶 ,制成了葡萄糖氧化酶电极。采用恒电位安培检测 ,葡萄糖浓度在0.05~4.75mmol/L范围内与响应电流成线性关系 ,检出限为0.02mmol/L ,灵敏度高达1.182μA·mmol -1·L ,响应时间为12s ;该酶电极不受抗坏血酸、尿酸等电活性物质的干扰 ,稳定性较好。该电极用于临床血糖的测定 ,回收率为96 %~110 % ,测定结果与酶偶联比色法结果一致 ,可用于临床糖尿病人血糖的检测。
A glucose oxidase electrode was prepared by immobilizing glucose oxidase(GOD)on a Prussian blue(PB)modified glassy carbon electrode,on which a thin layer of silica sol-gel was applied to fix the enzyme.For the determination of glucose,the amperometric method with constant potential was emˉployed.The electrode was found to be linear in the range of0.05~4.75mmol/L with a high sensitivity of1.182μA·mmol -1 ·L in view of the high electro-catalytic sensitivity of PB towards H 2 O 2 and the sol-gel immobilization as well.Its response time is12s,while its detection limit is0.02mmol/L gluˉcose.No interference from ascorbic acid and uric acid was observed.The enzyme electrode is quite staˉble.It has been applied to the clinical assay of glucose in human serum.The recovery was found to be in the range of96%~110%.The testing results of real samples are in satisfactory agreement with those of the spectrophotometric enzyme-coupling method.
