摘要
农杆菌介导的基因转化成功与否及效率与所选择的外植体类型有密切关系。报告以非洲菊叶柄基部的愈伤组织为起始材料,将其匀浆分散成小细胞团,并用该小细胞团为外植体进行农杆菌介导的基因转化,经GUS组织化学染色和抗生素筛选培养证明,用此体系可高效率地获得非洲菊转基因愈伤,该愈伤可能进一步再分化出转基因植株。
Agrobacterium-mediated genetic transformation is closely related to the type of explants. In this article, we use calli of Gerbera in vitro from grown shoots with petiole base as starting plant material for transformation. Calli were dispersed as micro-masses through blending . Histochemical analysis of GUS and section culture confirm that using the micro-masses as explants the transformed calli can be obtained with high efficiency.
