农杆菌介导的美洲商陆抗病毒蛋白基因对非洲菊遗传转化研究 被引量:4

摘要 以非洲菊叶柄基部的愈伤组织为材料,通过农杆菌介导法,建立高效稳定的遗传转化体系.结果表明,OD600值为0.3的菌液浓度,侵染时间8 min,36 h的共培养,延迟筛选2 d有利于获得较高的转化效率.通过农杆菌介导,持续筛选和PCR检测,美洲商陆蛋白(PAP)基因成功转入非洲菊幼苗. A highly efficient and stable Agrobacterium-mediated genetic transformation system for Gerbera jamesonii Bolus was established using the calli from the petiole base of the plant as the starting materials for transformation.The highest transformation frequency was obtained in this study with an OD600 of about 0.3 for Agrobacterium,an infection duration of 8 min,co-culture for 36 hours,and detention screening for 2 days.PCR proved that the pokeweed antiviral protein(PAP) gene had been successfully integrated into G.jamesonii Bolus.
