兔骨髓基质细胞同种异体移植修复骨缺损的实验研究 被引量:2

摘要 目的 :观察同种异体骨髓基质细胞移植治疗骨缺损的效果。方法 :取新生新西兰大白兔四肢长管骨骨髓 ,分离后加入条件培养液体外培养 ,冻存复苏的细胞以明胶海绵吸附后回植于异体兔的桡骨缺损处作为实验侧 ,对侧以明胶海绵吸附生理盐水作为对照侧。分别于术后 2、4、8、 1 2周取材 ,观察其修复骨缺损的能力和免疫排斥反应。结果 :植入异体骨髓基质细胞明胶海绵复合体未见明显的免疫排斥反应 ;术后 1 2周实验侧全部骨性愈合 ,对照侧由纤维组织填充。结论 Objective: To observe the therapeutic effect of transplanted allogeneic bone marrow stromal cells (BMSCs) in bone repair. Methods: The bone marrow cells were obtained from long bone of new born Zelanian rabbits, the cells were isolated and cultured in vitro in conditioned medium, then the acquired cells which had propagated in vitro and undergone frozen storage and resuscitation and gelatin sponge were made into a complex.The complex was then transplanted into the radius bone defect of allogeneic rabbit as the experiment side. Gelatin sponge absorbing physiological Saline was used in the contralateral side as the control side. The rabbits were sacrificed 2,4,8 and 12 weeks after implantation to observe healing states and immunological rejection reaction between the tow sides. Results: The transplantation of the complex of the allogeneic bone marrow stromal cells and gelatin sponge causes no obvious immunological rejection reaction. Twelve weeks after implantation,in the experimental side,all specimens had complete bony union;and in the control side,the specimens were filled with fibrous tissues. Conclusion:The result suggested that transplantation of the allogeneic bone marrow stromal cells could repair the bone defect effectively.
