涎腺肿瘤中P16蛋白和P16 mRNA的表达及意义

摘要 目的 :探讨P16蛋白和P16mRNA在涎腺良、恶性肿瘤中的表达及其临床病理意义。方法 :应用免疫组化和原位杂交方法检测了 5例正常涎腺和 6 4例涎腺肿瘤标本中P16蛋白和P16mRNA的表达 ,并分析了P16蛋白和P16mRNA表达与患者临床病理参数的关系。结果 :P16蛋白在涎腺良、恶性肿瘤中的阳性表达率分别为 73.9%(17 2 3)和 4 3.9% (18 4 1) (P <0 .0 5 ) ;P16mRNA在良、恶性涎腺肿瘤中的阳性表达率分别为 82 .6 % (19 2 3)和 4 8.8%(2 0 4 1) (P <0 .0 1) ;P16蛋白和P16mRNA表达与患者性别、年龄、肿瘤大小、部位均无相关性 ,但与恶性涎腺肿瘤的分化程度及是否有淋巴结转移密切相关 (P <0 .0 5 )。P16蛋白表达和P16mRNA表达具有相关性 (P <0 .0 5 )。结论 :涎腺肿瘤中P16蛋白和P16mRNA均有表达下降 ; Objective: To study the expression of P16 protein and mRNA in salivary gland tumors and its clinicopathological significance. Method: SABC immunohistochemical method and in situ hybridization method were used to investigate the expression of P16 protein and mRNA in 5 cases of normal salivary gland tissues and 64 cases of salivary gland tumors. The relationship between P16 protein and mRNA expression and such parameters as patient sex, age, tumor location, size, metastases and differentiation of malignant tumors was analyzed using x 2 test. Results: Positive rate of P16 protein expression was 73.9%(17/23) and 43.9%(18/41) in benign and malignant salivary gland tumors respectively (P<0.05); Positive rate of P16 mRNA expression was 82.6%(19/23) and 48.8%(20/41) in benign and malignant salivary gland tumors respectively (P<0.01) The P16 protein and P16 mRNA expression was not obviously correlated with patient sex, age, tumor size, location, but significant correlation was found between P16 protein and P16 mRNA expression and metastases and differentiation of malignant tumors (P<0.05). Conclusions: .Loss of P16 protein and P16 mRNA expression may play an important role in the tumorigenesis and development of salivary gland tumors.
