涎腺肿瘤端粒酶活性的表达及其临床意义的研究 被引量:1

摘要 目的 :检测端粒酶活性在良、恶性涎腺肿瘤中的表达 ,探讨其与涎腺肿瘤的性质、TNM分期、瘤体大小及淋巴结转移等临床参数之间的关系。方法 :采用TRAP法检测冻存的 87例良、恶性涎腺肿瘤标本的端粒酶活性。结果 :①涎腺恶性肿瘤端粒酶的阳性率显著高于良性肿瘤 ;良、恶性涎腺肿瘤的瘤旁组织中均无端粒酶活性表达。②端粒酶的阳性率与TNM分期无相关性。③瘤体长径 <2 0mm组端粒酶阳性率显著低于≥ 2 0mm组。淋巴结转移阴性组的端粒酶阳性率显著低于淋巴结转移阳性组。结论 :端粒酶活性的表达为恶性肿瘤的重要标志 ,端粒酶活性与涎腺肿瘤的增殖和淋巴结转移有相关性。检测涎腺肿瘤端粒酶的活性 ,有助于早期鉴别其良、恶性 ,为制定临床治疗方案提供理论依据 ,可帮助预测恶性涎腺肿瘤患者的预后。 Objective:To detect telomerase activity in 87 benign and malignant salivary gland tumors and their adjacent no-tumor tissues,compare the telomerase expression in benign neoplasma with malignant lesions,research the correlation between telomerase activity and proliferation,metastasis of malignant salivary gland tumors.Methods:Telomerase activity was detected by a telomeric repeat amplification protocol ( TRAP) assay.Results:①Positive rate of telomerase in malignant salivary gland tumors is significant higher than benign mass (P<0.01),telomerase activity in the adjacent no-tumor tissues of benign salivary gland mass and malignant ones is negative.②Positive rate of telomerase in TNM stage Ⅱ?Ⅲ?Ⅳ malignant salivary gland is significant higher than stage Ⅰ (P<0.05).There is no significant correlation between telomerase positive rate and TNM stages.③Telomerase positive rate in <20mm tumor group was significant lower than ≥20mm group(P<0.01) .④Node negative of malignant salivary gland tumor group was significant lower than node positive group (P<0.01).Conclusions:The expression of telomerase was closely related with the proliferation and metastasis of malignant salivary gland tumors. Telomerase may be a specific marker used to distinguish malignant salivary gland tumors from their benign counterparts and predict the prognosis of salivary gland malignancies.
