陆地棉分子标记辅助轮回选择聚合育种——Ⅰ.群体创建与基础群体的评价 被引量:6

Pyramiding Breeding by Marker-assisted Recurrent Selection in UplandCotton Ⅰ.Populations Construction and the Basic Population Evaluation

摘要 以抗黄萎病轮回选择基因库g2中的不育株为主体亲本,分别与高强纤维品系P7235、抗黄萎病种质P60182转Bt基因抗虫棉山西Bt(SHXBt)和中心Bt(ZHXBt)杂交后再进行聚合杂交,合成用于分子标记辅助选择的基础群体C0。随后进行分子标记辅助选择和表型选择,共获得P1、M1、MP1、P2、M2和MP2等6个选择群体。分析C0群体中的3种分子标记,发现基础群体C0的遗传基础是平衡的。将C0、g2与对照SU12进行比较,C0群体的抗虫性和各纤维品质性状均显著或极显著优于g2和SU12,早熟性和产量性状不显著低于g2与SU12;多数性状在3个群体之间的样本方差存在差异,而且以C0群体的选择潜势较大。比较了各性状在可育株与不育株之间的差异,明确了进行进一步分析的材料基础。 The initial population (C0) for marker-assisted recurrent selection was constructed by crossing male sterileplants segregated from a gene pool (g2) for resistance to Verticillium dahilae before with other 4 parents such as high fiberstrength germplasm line P7235, Verticillium dahilae-resistant line P60182, and transgenic Bt lines SHXBt and ZHXBt. Twocycles of marker-assisted selection, phenotypic selection, and marker/phenotype concurrent selection produced the totalsix populations named as M1, P1, MP1, M2, P2 and MP2. It is indicated that the genetic basis of C0 population was balancesince the segregation ratios of Bt gene, resistance to Kanamycin (Km) gene and a RAPD marker (St) flanking a major QTL(QTLFS-1) for fiber strength were normal. The means of fiber qualities and percentage of plants for resistance to bollwormof C0 population were significantly higher than that of g2 and SU12. The phenotypic variances of the two traits did not havethe homogeneity among the three populations, and the C0s were bigger than g2s and SU12s. The means of yield and itscomponents, and maturity of C0 population were a little bitter lower than that of g2 and SU12. And phenotypic variances formost of the traits were homogeneity in the three populations. So the relative selection potential for the multi-traits wasgreater in C0 population than in g2 and SU12. The differences of different traits between fertile and sterile plants wereanalyzed and the basic materials for further analysis were made clear and definite.
