摘要
目的 研究盐酸小檗碱对人早幼粒白血病 HL- 6 0细胞增殖与分化的影响。 方法 应用锥虫蓝排染法、生长曲线测定法、克隆形成实验检测药物对 HL- 6 0细胞生长的影响 ;细胞分化根据细胞形态、硝基四氮唑蓝(NBT)还原能力、细胞表面标记的改变来判断 ;细胞周期变化用流式细胞仪测定。 结果 HL- 6 0细胞经小檗碱处理后生长明显受抑 ,IC50 为 5 .2 9(2 4 h) ,2 .6 5 (4 8h) ,0 .74 (72 h) m g/L。选择 1 ,2 ,4 ,8mg/L 小檗碱作用于 HL- 6 0细胞 ,动态观察发现 HL- 6 0细胞向成熟细胞分化 :细胞核变小 ,核浆比减少 ;NBT还原能力显著增强 ;CD1 1 b表达升高 ,4 mg/L 组 CD1 1 b阳性率高达 85 .1 %。细胞周期分析发现 ,小檗碱处理后细胞阻滞于 G0 /G1 期 ,S期细胞明显减少。 结论 盐酸小檗碱可抑制 HL- 6 0细胞的增殖 ,并诱导 HL- 6
Objective To study effects of berberine on the proliferation and differentiation of HL-60 cells. Methods Cell inhibitory effect were determined by trypan blue dye exclusion test, growth curve method and colony formation. Cell differentiation was analyzed by morphological changes, NBT reduction and cell surface antigen. Cell cycle was analyzed by flow cytometry. Results The growth of HL-60 cells was significantly inhibited by berberine. The IC-{50} to HL-60 cells of berberine was 5 29, 2 65, 0 74 mg/L after in vitro treatment for 24 h, 48 h, 72 h, respectively. 1, 2, 4, 8 mg/L berberine could induce differentiation of HL-60 cells: morphological evaluation indicated that the berberine-treated cells exhibited granulation appearance; NBT reduction was significantly increased; expression of CD-{11b} increased, and 85 1% of the HL-60 cells expressed CD-{11b} after treated with 4 mg/L berberine. Flow cytometry analysis showed that berberine arrested HL-60 cells in G-0/G-1 phase, the percent of the cells in S phase decreased. Conclusion Berberine can induce differentiation and growth inhibition of HL-60 cells.;
