Establishment of Genetic Transformation Regeneration System in Brassica juncea

摘要 以叶用和茎用芥菜的子叶、叶片及下胚轴为外植体 ,通过筛选 12种含不同种类和浓度激素的培养基 ,综合分析与确定影响芥菜转基因再生体系各因素的最佳值 ,以及对再生抗性苗的PCR、Southern、Northern及抗虫鉴定 ,筛选到了能稳定表达外源基因的转基因植株 。 Using hypocotyls, cotyledons and leaves as explants, 12 sorts of media containing different kinds and concentrations of hormones were studied to establish an efficient regeneration system. The optimum values of the factors affected mustard transgenetic regeneration system were determined. The regenerated plants were identified by PCR, Southern, Northern blot and insect-resistance test. The target plants with cpti gene activity were separated from the Kan-resistant regenerated plants. Therefore the perfect transgenetic regeneration system of mustard was established.
