转天麻抗真菌蛋白基因彩色棉新品系抗枯黄萎病研究 被引量:9

Study on Resistance to Fusarium Wilt and Verticillium Wilt for Transgenic GAFP Colored Cotton Lines

摘要 将天麻抗真菌蛋白基因 (GastrodiaAntifargalProtein ,GAFP)用花粉管通道法转化天然彩色棉主栽品种和高代系 ,转化后代经卡那霉素初筛、分子检测、综合农艺性状选择 ,获得整合了GAFP的彩色棉花新品系。对其进行了室内抗枯萎病性鉴定和田间人工病圃抗黄萎病性鉴定。结果表明 :彩B5 - 8相对病指 4 93,为高抗枯萎病类型 ,彩B5 - 3相对病指 7 75、彩B5 - 9相对病指 8 0 9,为抗枯萎病类型 ;彩B5 - 7枯萎病、黄萎病相对病指分别为 9 95和 19 13,既抗枯萎病又抗黄萎病 ,是一个兼抗类型 ,彩B5 - 2相对病指 19 6 9为抗黄萎病类型。 The gene of Gastrodia Antifargal Protein(GAFP) was introduced into localreleused colored cotton cultivars and potential lines in Xinjiang province by using pollen-tube pathway method.Through kanamycin check, PCR-southern blot test and agronomic character identifi-cation, the new lines of colored cotton which have been integrated GAFP gene were obtained. The resistance identification to Fusarium wilt in the green house and to Verticillium wilt in the diseased field nursery proved that: The lines of B5-8, B5-3, B5-9 were resistant to Fusarium wilt, the relative indexes of resistance were 4.93, 7.75 and 8.09 respectively. B5-2, its relative index of resistance was 19.69, was resistant to Verticillium wilt. And B5-7 was resistant to both Fusarium wilt and Verticillium wilt, Its relative indexes of resistance were 9.95 and 19.13 respectively.
