骨疏康对1型糖尿病大鼠体外培养的破骨细胞影响的初步探讨 被引量:4

摘要 目的 探讨骨疏康对1型糖尿病大鼠体外培养的破骨细胞影响。方法 用RANKL和M-CSF诱导1型糖尿病大鼠的股骨骨髓进行体外培养生成类破骨细胞样细胞(OLC)。实验分为正常对照组,1型糖尿病组,正常对照+骨疏康组,1型糖尿病+骨疏康组。骨疏康的浓度为20μg/ml。细胞培养7天后,贴壁细胞固定,抗酒石酸盐酸性磷酸酶染色,破骨细胞计数。结果 1型糖尿病大鼠与正常大鼠组的破骨细胞数比较无明显差别(P>0.05);经骨疏康干预后,1型糖尿病大鼠与正常大鼠的破骨细胞数都明显减少(P<0.01)。结论 1型糖尿病早期破骨细胞形成与正常对照组无明显差异。中药骨疏康明显抑制正常大鼠和1型糖尿病大鼠的破骨细胞形成。 Objective To study the effect of Gushukang on osteoclast cultured from type I diabetic rat in vitro. Methods Osteoclast - like cells were induced from bone marrow cells by RANKL and M - CSF in vitro. The rats were divided into 4 groups: control group, type I diabetes mellitus ( DM ) group, control + Gushukang group, DM + Gushukang group. The concentration of Gushukang solution is 20(μg/ml. The bone marrow cells were cultured in 24 wells plate within 7 days, thus stained by TRAP and osteoclast cells were counted. Results There was no significant different (P > 0. 05 ) between rats with type I diabetes mellitus and the control in the number of osteoclasts; the number of osteoclasts both in type I diabetes mellitus group and control group treated with Gushukang were obviously decreased ( P < 0. 01 ) . Conclusions 1. There was no significant different between the formation of osteoclasts cultured from the rats with type I diabetes mellitus and that from control rats in vitro. 2. Gushukang inhibit the formation of osteoclast cultured from the rats with type I diabetes mellitus and control rats in vitro.
