摘要
以西瓜栽培种中育 1号为材料对影响西瓜组织培养的多个因素进行了研究 ,建立了西瓜茎尖、子叶块的高频再生系统。结果表明无菌苗子叶呈淡绿色时不定芽诱导率最高 ,茎尖和子叶块适于直接诱导不定芽 ,其中茎尖的诱导率稍高于子叶块 ,MS +6 -BA 1 0 +IBA 0 5可较好的诱导子叶块分化不定芽 ,MS +6 -BA 1 0 +IBA 0 1对茎尖较为合适 ,低浓度的IAA有利生根 ,低温。
A high regeneration frequency system of watermelon was set up by using the watermelon cultivated species-ZhongYuYiHao. Results indicated that MS+6-BA 1.0+IBA 0.5 was the most suitable medium for cotyledon and MS+6-BA 1.0+IBA 0.1 for stem apex;the strectched shoot rooted easily in MS+IAA 0.2;low temperature and high humidity may be helpful for the survival of test-tube seedlings.
