摘要
研究了用芯片毛细管电泳-激光诱导荧光检测系统分离多种荧光素异硫氰酸酯(FITC)衍生氨基酸的实验条件.通过实验优化了夹流式进样的电压条件.采用以乙醇作有机添加剂的胶束毛细管电动色谱分离体系(50mmol/LSDS,15%乙醇,5mmol/LpH9.2的硼砂缓冲液),在72mm长的通道上实现了10种常见氨基酸的分离,一次分离的时间小于5min.
Separation of FITC-derived amino acids by chip-based micellar electrokinetic capillary chromatography with laser induced fluorescence detection was investigated. The condition of applied voltages for pinched sample injection was optimized. Organic additive ethanol was found effective to improve resolution. By using a separation medium containing 50 mmol/L SDS, 15 ehtanol,and 5 mmol/L borate buffer (pH9.2), ten types of FITC-derived amino acids were separated in less than 5 minutes on a glass chip with effective separation length of 72 mm.
