性激素水平对大鼠动脉壁雌激素受体含量的影响 被引量:2

Effect of Sexual Hormone Level in Blood on the Expression of Estrogen Receptor on the Wall of Ovariectomized Rats' Artery

摘要 目的 研究循环中性激素水平对大鼠动脉壁雌激素受体 (estrogenreceptor ,ER)含量变化的影响。 方法 将5 0只成年雌性大鼠随机分为 5组 :假手术对照组、去卵巢组、去卵巢 +雌激素组、去卵巢 +雌孕激素组、去卵巢 +雌孕雄激素组。用药后给予正常饮食 ,8周后处死大鼠。检测血清中雌二醇、孕酮、睾酮浓度及检测大鼠主动脉降段ER的含量。结果 正常雌性成年大鼠主动脉中存在ER ;去卵巢组大鼠动脉ER的含量显著低于假手术对照组 ;去卵巢 +雌激素组与去卵巢 +雌孕激素组及去卵巢 +雌孕雄激素组动脉ER ,3者都显著高于去卵巢组 ;去卵巢 +雌孕雄激素组的体重明显高于其他两治疗组。结论 去卵巢后大鼠主动脉的ER显著减少 ,补充雌激素或补充雌孕激素及雌孕雄激素能使去卵巢大鼠主动脉ER的含量达到正常水平 ,这可能是雌激素对绝经后妇女心血管保护作用的机制之一。雄激素可提高循环中雌激素的量 。 Objective To investigate the effect of sexual hormone level in blood circulation on the expression of estrogen receptor (ER) on the wall of ovariectomized rats' artery.Methods Fifty female rats were randomly divided into five groups:operation control(A);ovariectomy(B);ovariectomy plus estrogen supply(C);ovariectomy plus estrogen and progesterone supply(D);ovariectomy with estrogen,progesterone and testosterone(E).The rats were given normal diet and killed 8 weeks after treatment.Sexual hormone level in blood circulation and the expression of estrogen receptor in the artery of ovariectomized rats were measured.Results ER was found in the arteries of adult female rats(group A).The amount of ER in the artery of group B was less than that in groupA(-P-<0.01).The level of ER in the artery of group C,D and E were higher than that in group B.The weight of group E was higher than that in group C and D.The ER number on the wall of the artery of rats was significantly decreased after ovariectomy.Supplementation of estrogen or estrogen and progesterone or estrogen,progesterone and testosterone might maintain the ER amount in the wall of the artery of ovariectomized rats at normal level.Conclusion The results of this study might help to explain the beneficial effect of estrogen in preventing cardiovascular diseases in post menopausal women.Testosterone therapy can increase estrogen level in blood circulation,but its clinical application warrants further study.
