摘要
目的 研究交联琼脂包嵌凹凸棒微囊 (CAA)正常大鼠血液灌流中的血小板、白细胞及红细胞比积的变化 ,进一步认识该材料的体内血液灌流生物相容性。方法 以Wistar大鼠为实验对象 ,分别于第 1、3、5天重复血液灌流 3次 ,每次于灌流前及灌流 1h后取鼠尾血样 ,分别测试血小板、白细胞、红细胞比积。结果 经CAA血液灌流 1h后相应血小板、白细胞、红细胞比积无明显降低 (P >0 0 5 )。至 48h灌流时其血小板、白细胞有所降低 ,但差异不显著 (P >0 0 5 ) ,而红细胞比积降低较明显 (P <0 0 5 )。经灌流后的存活大鼠至 96h其血小板、白细胞基本恢复正常 ,红细胞比积仍有下降 ,但与 48h相比较明显减缓。结论 新型血液净化吸附剂CAA血液灌流对动物血液有形成分的影响是暂时的 ,该材料具有较好的血液相容性 ,多次血液灌流对动物存活并无明显影响。
Objective To investigate the blood compatibility of cross linked agar beads entrapped attapulgite clay(CAA) by observation of the changes of blood platelet count, white blood cell count and hematocrit in normal Wistar rats during hemoperfusion. Methods Repeated hemoperfusion was performed on Wistar rats on the first day, the third day and the fifth day respectively. At 1 h before and after hemoperfusion each time, blood platelet count, white blood cell count and hematocrit in the blood samples collected from the Wistar rat tails were observed. Results No significant decrease in blood platelet count, white blood cell count and hematocrit was found at 1 h after CAA hemoperfusion( P >0.05). Blood platelet count, white blood cell count decreased slightly ( P >0.05) but hematocrit fell notably( P <0.05) at 48 h after hemoperfusion. At 96 h after hemoperfusion, blood platelet count and white blood cell count in the survival rats restored to normal but decrease in hematocrit was also found, though the decrease was slower than that at 48 h after hemoperfusion. Conclusion CAA, as a new kind of blood purification adsorbent, has no significant effect on the survival rate of rats due to the good blood compatibility though it has temporary effect on the formed blood elements.
