人肝癌组织中乙酰肝素酶基因的克隆与序列分析 被引量:1

摘要 目的 :研究乙酰肝素酶在人肝癌细胞生长和转移中的作用 ,并自人肝癌组织中克隆该酶cDNA全长编码片段 .方法 :提取人肝癌细胞总RNA ,以RT PCR方法获取了乙酰肝素酶cDNA片段 ,将其与pGEM TEasy载体连接 ,进行克隆及酶切鉴定 .结果 :序列分析表明 ,与GenBank中的人乙酰肝素酶全长cDNA序列比较 ,仅 1 6个碱基不同 ,同源性为0 .991 .结论 :自人肝癌组织中成功克隆了乙酰肝素酶cDNA全长编码片段 . AIM: To study the role of heparanase in the growth and metastasis of human hepatocelluar carcinoma (HCC), and to colone heparanase cDNA gene from HCC. METHODS: The cDNA fragment encoding heparanase was isolated by RT PCR method with the total RNA extracted from human hepatoma, linked into pGEM T easy vector, then amplified, cloned, finally the recombinant plasmids was identificated by Electrophoresis. RESULTS: The result of sequencing indicated that the sequences of our cDNA was identical to the corresponding parts of the mRNA for human heparanase in GenBank except 16 nucleotides, and with a homoloty of 0.991. CONCLUSION: Human heparanase cDNA gene is successfully cloned from HCC.
