摘要
目的:探讨乙酰肝素酶在食管鳞状细胞癌(ESCC)组织中的表达及其与ESCC临床病理特征间的关系。方法:应用RT—PCR和免疫组化方法检测55例ESCC术后组织标本及相应癌旁组织中乙酰肝素酶的表达,分析乙酰肝素酶表达与ESCC临床病理特征的关系。结果:ESCC组织中存在乙酰肝素酶基因的表达,并定位于肿瘤细胞质和(或)细胞膜中。55例ESCC组织中有40例乙酰肝素酶基因mRNA表达阳性(40/55.72.7%),癌旁组织有19倒表达阳性(19/55,34.5%),两组间差异有显著性(P〈0.01)。免疫组化染色ESCC组织中乙酰肝素酶阳性率63.6%(35/55),癌旁组织阳性率21.8%(12/55),两组间差异有显著性(P〈0.01)。乙酰肝素酶基因mRNA和蛋白质的检测均显示有淋巴结转移的癌组织乙酰肝素酶表达高于无淋巴结转移组,TNM分期Ⅲ~Ⅳa的癌组织表达高于Ⅰ~Ⅱ组,均有统计学意义。结论:ESCC组织乙酰肝素酶基因及蛋白表达高于正常组织,并且与ESCC的转侈和进展有密切关系。
Objective:To investigate the expression of heparanase in esophageal squamous cell carcinoma (ESCC) tissue and the relationship between heparanase expression and clinical pathological features. Methods: Expression of heparanase gene in 55 surgically resected ESCC tissues and adjacent normal esophageal tissues were detected by RT-PCR assay and immunohistochemical staining. Then the relationship between heparanase expression and ESCC clinical pathological features was explored. Results: There were heparanase gene and protein expressions in ESCC tissues. Heparanase protein was located in tumor cell cytoplasm and/or cell membrane. The rate of heparanase mRNA expression in cancer tissues (40/55,72.7 % ) was significantly higher than that in adjacent normal tissues (19/55.34. 5%) ( P 〈0. 01). Heparanase expression was correlated to lymphatic metastasis (P〈0.01) and TNM stages( P 〈0. 05). The rate of heparanase protein expression in cancer tissues(35/55,63. 6%) was significantly higher than that in adjacent normal tissues( 12/55,21.8% )( P〈0.01 ). Heparanase expression was higher in cancerous tissues with lymphatic metastasis than that without lymphatic metastasis ( P 〈0.01 ). Heparanase expression was higher in TNM Ⅲ-Ⅳ stages of cancerous tissues than that in Ⅰ-Ⅱ stages ( P〈0.01 ). Conclusions: Heparanase is over-expressed in cancerous tissues compared with adjacent tissues. Expression of heparanase is correlated to metastasis and development of esophageal squamous cell cancer.
