实验性局部脑缺血及其组织病理学观察

摘要 本研究共用威斯特大鼠40只,以闭塞大脑中动脉(MCA)的方法复制局部脑缺血的实验模型。本文描述了闭塞MCA的手术方法,并对MCA闭塞后脑组织病理学,梗塞面积及电子计算机断层扫描(CT)结果进行了探讨。结果表明,本方法复制的局部脑缺血模型死亡率低,重复性高。脑缺血后脑组织病理学改变有神经细胞水肿、线粒体固缩、细胞器崩解,核膜破裂,细胞坏死等,而小胶质细胞的噬节细胞现象和大量格子细胞的形成为最显著的特点之一。MCA闭塞后脑梗塞的面积轻恒定,但CT扫描未见缺血性改变,由于该研究用小啮齿动物,采源便宜、手术操作较简便,且术后存活卒高;可供临床研究脑缺血时形态、机能及代谢的改变作参考。 40 wistar rats were used for this study, and an experimental model of local cerebral ischemia was established by the middle cerebral artery (MCA) occlusion. In this paper, authors described a surgical method for the occlusion of MCA and evaluated the brain histopathology, brain perfusion, brain infarct area and transverse computed tomography (CT) scan after the occlusion. The results showed that this local cerebral ischemia model had a low mortality rate and an excellent reproduction mte. The histopathologic changes of the brain after ischemia involved neuron swelling, mitochondrial pyknosis and other organelles crumbling. And the neuronophagy of the microglia and a lot of gitter cells formation were the most striking feature. The infarct area was rather identical but the ischemia alteration could not be differentiated precisely by CT. The small rodent as model is convenient because the operative procedure is simple, the post-operative mortality is almost negligible and survival for a few weeks is easy to achieve.Therefore, this model is available for the clinic to study the morphological, functional and metabolic changes during cerbral ischemia.
